不同醇沉浓度对广西莪术多糖含量的影响 被引量:16

摘要 目的优选广西莪术多糖水提醇沉浓度,为广西莪术的开发和利用提供理论基础。方法以广西莪术多糖作为指标性成分,采用热水浸提法提取多糖,以含醇量为65%,75%,85%,95%优选广西莪术多糖提取液的醇沉浓度,采用苯酚-硫酸比色法测定多糖含量。结果葡萄糖在2.857~14.286μg/ml范围内与吸光度呈良好的线性关系,r=0.999 2;平均回收率为99.94%,RSD=0.20%(n=6)。65%、75%、85%、95%醇沉浓度下测得的广西莪术多糖分别为6.698,8.499,6.855,6.170μg/ml,RSD分别为3.43%,2.10%,3.55%,6.47%(n=3)。结论不同醇沉浓度对广西莪术多糖含量影响较大,广西莪术多糖最佳醇沉浓度为75%。 Objective To optimize the concentrations of alcohol in precipitation method and provide a theoretical basis for the de- velopment and use of Curcuam kwangsiensis. Methods The Curcuam kwangsiensis polysaceharide (CKP) was used as an index component, the CKP was extracted by hot water extraction method, the concentrations of alcohol in precipitation method was opti- mized by the concentrations of alcohol of 65% , 75% , 85% and 95% , the CKP content was detected by phenol - sulfuric acid method. Results The concentration glucose within the range of 2. 857 - 14. 286 μg,/rnl was linear with the absorbance, r = 0. 999 2; the average recovery was99.94%, RSD = 0.20% (n = 6). The CKP content was respectively 6. 6981.μg/ml, 8. 499p, g/ml, 6. 855μg/ml and 6. 170μg/m1, the RSD was respectively 3.43% , 2.10% , 3.55% and 6.47% (n = 3) in the al- cohol precipitation concentration of 65%, 75%, 85% and 95%. Conclusion The alcohol precipitation concentration has great impact on the CKP content, the best concentration of alcohol in precipitation method is 75%.
