摘要
目的探讨姜黄素(curcumin,CMN)抗大鼠对比剂肾病(CIN)的作用及机制。方法雄性SD大鼠24只,随机均分为对照组(A组)、单纯泛影葡胺组(B组)、泛影葡胺+CMN组(C组)、泛影葡胺+CMN+锌原卟Ⅸ(ZnPPⅨ)组(D组)。复制对比剂肾病模型并分别给予不同的药物干预,测定血清肌酐(Cr)水平及肾组织中血红素氧合酶-1(HO-1)表达量与活性、白细胞介素-6(IL-6)、单核细胞趋化蛋白-1(MCP-1)、丙二醛(MDA)水平,观察肾组织形态学改变。结果与A组相比,B、C、D组各项指标均有明显改变(P<0.05),而C组与B组和D组相比,HO-1表达量、HO-1活性明显升高(P<0.05),血清Cr、IL-6、MCP-1、MDA水平则明显降低(P<0.05),肾损伤程度明显减轻;D组HO-1活性受到显著抑制,其血清Cr、IL-6、MCP-1、MDA水平及肾损伤程度均明显高于其他各组。结论 CMN可诱导HO-1表达,并通过HO-1介导的抗炎症和抗氧化应激机制防止CIN,保护肾脏功能。
【Objective】 To explore the protective effects and mechanisms of curcumin(CMN) on the contrast-induced nephropathy(CIN).【Methods】 24 male SD rats were divided randomly into four groups: control group(Group A),diatrizoate(DTZ) group(Group B),DTZ+CMN group(Group C),DTZ + CMN + zinc protoporphyrinⅨ group(Group D).Rat's CIN model was established for different drug intervention,the serum creatinine(Cr) level was measured,the heme oxygenase-1(HO-1) expression and its activity as well as the content of interleukin-6(IL-6) and monocyte chemoattractant protein-1(MCP-1) and malondialdehyde(MDA) in renal were detected,the renal morphological change was also observed.【Results】 Compared with group A,all the indicators in the other groups had changed significantly(P 0.05 respectively).In group C,The HO-1 expression and its activity were significant higher while serum Cr,IL-6,MCP-1,MDA level were significant lower than that in group B and group D(P 0.05 respectively),the renal tissue damage of group C was also reduced significantly than that of group B and group D.In group D,the HO-1 activity was significantly inhibited while the serum Cr,IL-6,MCP-1,MDA level and the renal tissue damage were significant higher than that of the other groups.【Conclusion】 CMN could induce HO-1 expression and protect against CIN through anti-inflammatory and anti-oxidative stress mechanisms mediated by HO-1.
