摘要
目的探讨系统性红斑狼疮(SLE)患者外周血T淋巴细胞激活状态及OX40、OX40LmRNA表达水平。 采用双色流式细胞仪测定SLE患者和健康人外周血CD38CD3分子表达,采用荧光定量逆转录聚合酶链反应(RT—PCR)测定外周血单一核细胞(PBMC)中OX40、OX40LmRNA表达水平。结果SLE患者外周血CD38CD3分子表达水平显著高于健康人(t=3.12,P〈0.05)。活动期SLE患者PBMCOX40、OX40LmRNA水平显著高于非活动期患者和健康人(F=4.13,3.12,P值均〈0.01)。OX40mRNA水平与SLE疾病活动指数呈正相关(r=0.35,P〈0.05)。结论T淋巴细胞异常活化及OX40、OX40L mRNA表达水平上调在SLE的发生、发展中可能起一定作用。
Objective To evaluate the activation status of T lymphocytes and to measure the expression of OX40 and OX40L mRNA in peripheral blood from patients with systemic luptis erythematosus (SLE). Methods Peripheral blood was collected from 30 patients with SLE and 15 healthy controls. The expression of CD38 and CD3 on peripheral blood T lymphoeytes were quantified by using a two-color fluorescence activated cell sorter. Fluorescence-based quantitative reverse transcription PCR was performed to determine the expression levels of OX40 and OX40L mRNA in peripheral blood mononuclear cells (PBMCs). Results The coexpression levels of CD38 and CD3 were significantly increased in patients with SLE compared with healthy controls (t = 3.12, P 〈 0.05). Increased expression levels of 0)(40 and OX40L mRNA were observed in patients with active SLE compared with those with inactive SLE and healthy controls (F= 4.13, 3.12, both P〈 0.01). The level of OX40 mRNA was positively correlated with SLE disease activity index (SLEDAI) score in the patients (r = 0.35, P 〈 0.05). Conclusion Abnormally activated T lymphoeytes and upregulated expression of OX40 and OX40L mRNA may play a certain role in the initiation and progression of SLE.
