体外感染细胞表达HCV-NS5和复制HCV RNA的实验研究

摘要 为了研究 HCV- NS5 和 HCV RNA在体外感染细胞培养系统中的表达和复制情况 ,选用 HCV RNA水平为2× 10 4拷贝 / m l的阳性血清感染人肝癌细胞株 (Hep G2细胞株 )。应用胶体金标免疫电镜法和原位杂交法检测 HCV在被感染细胞中 NS5 蛋白表达和 RNA复制的情况。结果在细胞浆内有电子密度大的金标颗粒的吸附 ,在感染后的第 1至第 9代细胞中均有发现。感染后的细胞超微结构完整 ,未发现 HCV颗粒。在感染后的第 1至第 7代细胞内出现特异性杂交信号 ,HCV RNA阳性物呈蓝紫色细小颗粒分布在细胞浆和细胞核内。因此 ,HCV能够在 Hep G2细胞中表达 NS5抗原和复制 RNA,而细胞超微结构完整。 In order to evaluate the expression of HCV non structure 5 antigen (HCV NS5) and replication of HCV RNA in the culture system of the infectious cells in vitro, a human HepG2 cell line was incubated with positive serum containing HCV RNA of 2×10 4 copy/ml. The gold labeled coloid electron microscopy and in situ hybridization were employed to detect the expression of HCV NS5 protein and the replication of HCV RNA in the infected cells. In the cytoplasm, the adsorptions of gold labeled particles with high electronic density were found in the cells of generation 1 to generation 9 after infection. The ultrastructure of the cells after infection was intact and no adsorptions of HCV particles were found. The special hybridization positive signals appeared in the cells of generation 1 to generation 7 after infection. HCV RNA positive signals were shown as blue purple small graunles in the cytoplasm and on the membrane. It was suggested that HCV could expressed NS5 antigen and replicated RNA in the infected HepG2 cell line. The ultrastructure of cells was intact.
