摘要
建立了一种功能性饮料11种水溶性维生素(维生素B1、维生素B2、吡哆胺、吡哆醛、吡哆醇、泛酸、烟酸、烟酰胺、生物素、叶酸、维生素B12)高效液相色谱法高通量检测方法。采用C18反相色谱柱(150×4.6μm,5μm),以甲醇和磷酸二氢钾为流动相梯度洗脱,以DAD和荧光检测器进行串联检测,流动相流速为0.6mL/min,进样量10μL。分析了检测波长,流动相对方法的影响,本方法检出限为0.015mg/L~0.4mg/L,平均加标回收率为93%~107%,相对标准偏差为0.8%~3.7%。该方法重现性好、灵敏度高,是一种功能饮料中水溶性维生素检测的好方法。
A detection was developed for the determination of Water-soluble Vitamins in functional beverage by HPLC with fluorescence detector and DAD detector. The quantitative method was external standard method. Gradient elution was used for the baseline separation of the eleven water-soluble in 30 minutes. The optimized chromatographic conditions were : analytical column : Diamonsil C18(150×4.6μm,5um) ; column temperature:ambient;mobile phase with gradient elution:0.05mol/L potassium dihydrogen phosphate buffer/methanol from 96/4 to 60/40;flowrate:0.6mL/min. The detection limit of the method was from 0. 015mg/L to 0. μ mg/L. The mean recoveries were in the range of 93% and 107%,and accordingly the relative standard deviation ranged from 0.80/oo to 3.7 %. This method is repeatable, sensitive and accurate.
