再灌注前干预肥大细胞功能对肠缺血再灌注氧化损伤的影响

Effect of Intervention with Mast Cell Function before Reperfusion on Intestinal Ischemia-reperfusion Injury and Oxidative Stress

摘要 【目的】探讨再灌注前干预肥大细胞功能对肠缺血再灌注氧化损伤的影响。【方法】48只SD大鼠随机分为6组:S组(假手术组)、M组(缺血再灌注模型组)、C组(缺血再灌注+色甘酸钠)、CP组(缺血再灌注+Compound 48/80)、P组(缺血再灌注+鱼精蛋白)、K组(缺血再灌注+酮替芬)。复制大鼠肠缺血再灌注损伤模型,CS、CP、P、K组在再灌注前5 min分别经尾静脉给予色甘酸钠25 mg/kg、Compound 48/80 0.75 mg/kg、鱼精蛋白2.5 mg/kg、酮替芬1 mg/kg,S、M组分别给予等量的生理盐水。再灌注4 h后处死大鼠取小肠组织。观察各组肠黏膜病理变化及Chiu’s评分;免疫组化计算IMMC数量;测定小肠MDA含量、SOD活性和NO含量。【结果】与S组相比,M组和CP组的Chiu’s评分和IMMC计数均明显增高(P<0.05);与M组相比,C、P、K组的Chiu's评分和IMMC计数明显降低(P<0.05),而CP组的Chiu’s评分明显增高(P<0.05)。【结论】再灌注前通过抑制肥大细胞功能明显减轻氧化应激,对肠缺血再灌注损伤具有良好的保护作用。 [ Objective ] To explore the effect of intervention with mast cell function before reperfusion on intestinal ischemia- reperfusion and oxidative stress. [Methods]A total of 48 SD rats were randomly divided into 6 groups with 8 rats each: Sham operated group (S group), Intestinal ischemia reperfusion group (M group), and Cromolyn Sodium group (CS group, 25 mg/kg), Protamine group (P group, 2.5 mg/kg), Ketotifen group (K group, 1 mg/kg), Compound 48/80 group (CP group, 0.75 mg/kg). Model group (M group) was induced by clamping the superior mesenteric artery for 75 rain followed by reperfusion for 3 hours. The above agents were, respectively, intravenously administrated 5 min before reperfusion. Rats were then sacrificed and intestinal issues were harvested for histologic assessment. The mast cell count were analyzed by immunohistochemical detection, and the contents of MDA, SOD, and NO were detected. [Results] The Chiu's score and mast cell counts of group M and CP were significantly more than the group (both P 〈 0.05). Compared with M group, the Chiu's score and mast cell counts were less than in group C, P, and K, while the Chiu's score and mast cell counts were more in CP group (all P 〈 0.05). [Conclusion] Intervention with mast cell function before reperfusion may reduce intestinal ischemia-reperfusion injury and oxidative stress.
