摘要
目的观察白念珠菌氟康唑耐药株和敏感株中锌簇转录因子编码基因UPC2,TAC1,MRR1和相关耐药基因CDR1,CDR2,MDR1,FLU1和ERG11的表达情况。方法抽提白念珠菌氟康唑耐药株组和敏感株组的总RNA,逆转录合成cDNA,采用实时定量PCR法检测相关基因的表达情况。结果白念珠菌耐药株组UPC2和CDR2基因的表达(7.22±4.21和2.45±1.37)均高于敏感株组(1.05±0.05和1.03±0.09),差异均有显著的统计学意义(P均<0.05);但是,TAC1(1.46±0.86和1.05±0.12)和MRR1基因的表达(0.72±0.58和1.09±0.11)差异均无统计学意义(P均>0.05)。结论 UPC2和CDR2基因的高表达与氟康唑耐药有关,锌簇转录因子在耐药中的作用有待于进一步研究。
Objective To explore the relationship between fluconazole resistance and expression of C. albicans genes encoding transcription factors UPC2 ,TAC1, MRR1 and drug resistance-related genes CDR1, CDR2, MDR1, FLU1, ERG11. Methods The total RNA was extracted from fluconazole-resistant and-susceptible C. albi- cans isolates and cDNA was synthesized. The expression of drug resistance-related genes was then detected by quantitative real-time PCR. Results Higher expression of UPC2 ( 7.22 ± 4.21 ) / CDR2 ( 2.45 ± 1.37 ) gene was observed in the flueonazole-resistant isolates than that in the fluconazole-suseeptible isolates. The difference was statistically significant ( all P 〈 0.05 ). However, the expression of TAC1 ( 0.46 ± 0.86 and 1.05 ± 0.12)/MRR1 (0.72 ± 0.58 and 1.09 ± 0.11 ) was no statistical difference ( all P 〉 0.05 ). Conclu- sion It is likely that the over-expression of UPC2 and CDR2 gene is relevant to fluconazole resistance in C. albicans. The function of Zinc cluster transcription fators in drug resitance needs to be further studied.
