苜蓿素对人直肠癌SW1116细胞增殖和凋亡的作用及其机制 被引量:11

摘要 本文探讨苜蓿素对人直肠癌SW1116细胞增殖和凋亡的作用及其机制。采用MTT法检测苜蓿素对SW1116细胞生长的抑制作用;AO/EB染色后观察凋亡细胞形态;流式细胞术检测对细胞周期的影响;DNA片段化分析对细胞凋亡的作用;Western blot检测对Bcl-2和Bax蛋白表达的影响。结果显示,苜蓿素可明显抑制SW1116细胞的增殖,呈剂量依赖性;AO/EB染色观察到给药组出现细胞凋亡特征;苜蓿素可阻滞细胞于G0/G1期和增加SW1116细胞凋亡率;凝胶成像分析仪检测到典型的DNA阶梯状条带;苜蓿素可呈剂量依赖地减弱Bcl-2和增强Bax蛋白表达。上述结果表明,苜蓿素可显著抑制人直肠癌SW1116细胞的增殖,使细胞阻滞于G0/G1期,并可诱导细胞凋亡,其机制可能与下调Bcl-2和上调Bax蛋白有关。 In this study, the effects and mechanisms of tricin on proliferation and apoptosis of human rectal cancer SW1116 cells were investigated. MTF assay was used to detect the growth inhibition rates of human rectal cancer SW1116 cells treated with tricin. The apoptosis morphology was observed after AO/EB staining. The cell cycle distribu- tion was measured by flow cytometry. The apoptosis was examined by DNA fragment analysis. The protein expressions of Bcl-2 and Bax were detected by western blot. MTT assay showed that tricin inhibited the growth of SW1116 cells in a dose-dependent manner. The typical apoptotie morphological changes were observed. Triein blocked SW1116 cells in G0/ G1 and increased the apoptosis rates of SW1116 cells. DNA ladder was observed in cells with the gel imaging analysator. Western blot showed that tricin decreased expression of Bel-2 and increased expression of Bax in a dose-dependent, manner. The results show that tricin could inhibit human rectal cancer SWl 116 cells proliferation and induce cell cycle block in G0/G1 phase and apoptosis. The down regulation of Bcl-2 and up regulation of Bax protein might be one of the mecha- nisms.
