结直肠癌中NGAL和NGALR的表达及其临床意义 被引量:12

Expression and clinical significance of NGAL and NGALR in colorectal carcinoma

摘要 目的探讨中性粒细胞相关载脂蛋白(neutrophil gelatinase-associated lipocalin,NGAL)及其膜受体(neutrophil gelatinase-associated lipocalin receptor,NGALR)在结直肠癌组织中的表达及与结直肠癌临床病理因素之间的关系。方法应用免疫组化SP法检测NGAL和NGALR在结直肠癌、结直肠腺瘤和癌旁正常组织中的表达,并用统计软件对数据进行分析。结果NGAL和NGALR在结直肠癌组织中的阳性率分别为49.0%、35.4%,明显高于结直肠腺瘤和正常黏膜组织。NGAL和NGALR的表达与肿瘤的浸润、远处转移和TNM分期均相关,差异均具有统计学意义(P<0.05),且NGAL/NGALR共同表达与结直肠癌的分化程度、浸润深度、远处转移和TNM分期相关(P<0.05)。结论 NGAL和NGALR的表达与结直肠癌的发生、浸润和转移相关,可辅助临床诊断,并有可能成为结直肠癌临床治疗的新靶点。 Purpose To investigate the expression of neutrophil gelatinase-associated lipocalin (NGAL) and its membrane receptor (NGALR) in colorectal carcinoma specimens, and to determine the relationship between expression of these two proteins and tumour progression. Methods Expression of NGAL and NGALR was evaluated by immunhistochemistry in nomal colorectal mucosa, adeno- mas and eolorectal carcinoma. Statistical analyses of the data were performed with SPSS software. Results Expression levers of NGAL and NGALR were both up-regulated in eolorectal carcinoma tissues which significantly higher than that in nomal colorectal mueosa and adenomas tissues. Expression of NGAL and NGALR was associated with invasion, distant metastasis and TNM stages ( P 〈 0. 05 ). Es- pecially, co-expression of NGAL and NGALR was significantly correlated with histopathology, distant metastasis and TNM stages in colorectal cancer (P 〈 0. 05). Conclusion The data demonstrates that up-regulations of NGAL and NGALR in colorectal carcinoma are both significantly correlated with unfavorable clinicopathological features. These findings suggest that NGAL and NGALR expression might be served as novel factors to help clinical diagnosis and potential therapeutic targets in colorectal carcinoma.
