巴戟天糖链对ECV304细胞增殖中BK_(Ca)通道的影响

摘要 目的观察巴戟天糖链(MOO)对ECV304细胞增殖中大电导钙激活钾通道(BKCa)通道的影响。方法以人脐静脉内皮细胞株(ECV304细胞)为研究对象,制备缺氧/复氧模型,采用流式细胞仪检测MOO对ECV304细胞的增殖效应;在+60mV指令电压刺激下,应用全细胞膜片钳技术研究MOO对ECV304细胞增殖中BKCa通道电流密度的影响。结果与模型组相比,MOO大剂量组(0.45 g.L-1)与麝香保心丸组均促进缺氧/复氧损伤后ECV304细胞增殖,差异有显著性(P<0.05)。全细胞膜片钳实验提示:MOO三个剂量组均能增加BKCa通道的电流密度,差异有显著性(P<0.05),其中大剂量MOO组最为明显。结论 MOO可通过激活ECV304细胞BKCa通道保护受损细胞并促进其增殖。 Objective To observe the effect of morindae officinalis oligosaccharide(MOO) on large conductance calcium- activa- ted potassium channel (BKca) in ECV304 cell proliferation. Methods ECV304 cells were induced by hypoxia - reoxygenation (H/R) ;cell cycle distribution was examined by with flow cytometry(FCM) , channel activities of BKCa was recorded from patch clamp experiment under the electrical stimulation at + 60mV. Results MOO nigh dosage (0.45g L-1 ). and Shexiang Baoxin Wan increased the index of G2/S% (P 〈 0.05). Compared with model group, the current densities of BKca channel increased sig- nificantly in MOO different dosages (0.45g L-1. 0.15g L-1 and 0.05g L-1 ) ( P 〈 0.05 ). Conclusion MOO can Stimulate cell proliferation of ECV304 in hypoxia - reoxygenation injury. The mechanism may be related to activating large conductance cal- cium - activated potassium channel ( BKCa ) activities.
