摘要
目的:探讨宁神温胆汤对精神分裂症模型鼠海马神经细胞凋亡基因Caspace-3的表达影响。方法:将40只SD大鼠随机分为4组:正常组、模型组、氯丙嗪组、宁神温胆汤组。宁神温胆汤组于造模前采用连续灌胃给药,每次给药20 mL.kg-1(相当于灌胃20 mg.kg-1);氯丙嗪组先连续灌胃等容量生理盐水14 d,1次/d,最后7 d腹腔注射氯丙嗪4.5 mg.kg-1(按体表面积计算,相当于正常人用量);正常组和模型组用等容量生理盐水灌胃,灌胃1次/天,共21 d。除正常组外,其它3组大鼠最后一天给药1 h后,于左侧腹腔注射阿朴吗啡盐酸半水,按浓度为2 mg.kg-1体重(注射量为0.1 mL/20 g)的标准注射,诱发SD大鼠精神分裂症模型,观察15 min后断头处死。结果:模型组大鼠海马区Caspase-3蛋白表达的平均光密度明显高于正常组,平均灰度值明显低于正常组,差异具有统计学意义(P<0.01)。氯丙嗪组、宁神温胆汤组大鼠海马区Caspase-3蛋白表达的平均光密度明显低于模型组,平均灰度值明显高于模型组,差异具有统计学意义(P<0.05);宁神温胆汤组、氯丙嗪组两组组间差异有统计学意义(P<0.05)。结论:模型组大鼠海马Caspase-3蛋白表达较空白对照组明显增高;宁神温胆汤治疗后大鼠海马区Caspase-3蛋白表达明显降低。说明宁神温胆汤能调控精神分裂症模型鼠海马神经细胞凋亡基因Caspase 3表达,即抑制Caspase-3表达,明显抑制神经细胞凋亡,但宁神温胆汤的作用较氯丙嗪温和。
Objective:To investigate the influence of Ningshen Wendan decoction on Caspace-3 expression of hippocampal neuronal apoptosis in schizophrenia model rats.Methods:40 SD rats were randomly divided into four groups:normal group,model group,Ningshen Wendan decoction,Chlorpromazine group.Before modeling rats in Ningshen Wendan decoction group was given the continuous intragastric administration of each dose of 20 mL·kg^-1equivalent to the intragastric administration of 20 mg·kg^-1.Chlorpromazine group was given first intragastric administration of normal saline for 14 days,once a day,in the last 7 days was given intraperitoneal injection of Chlorpromazine at a dose of 4.5 mg·kg^-1according to body surface area,which is equivalent to the normal dosage),normal and model groups were given the capacity of saline intragastric administration,once a day,totally lasting 21 days.In addition to the normal group,on the last day of administration in the other three groups,1 h after administration,left intraperitoneal injection with Apomorphine Hydrochloride Hemihydrate at a concentration of 2 mg·kg^-1body weight(injection volume for 0.1 mL/20 g)to induce schizophrenia model,the rats were decapitated 15 min after observation.Results:The mean optical density of Caspase-3 protein expression in rat hippocampus of model group was significantly higher than that of the normal group,the average gray value was significantly lower than that of the normal group,with a statistically significant difference(P0.01).The mean optical density of Caspase-3 protein expression in Chlorpromazine group and Ningshen Wendan decoction group was significantly lower than that of the model group,the average gray value was significantly higher than that of the model group,with a statistically significant difference(P0.05);the difference between the two groups of Chlorpromazine group and Ningshen Wendan decoction group was statistically significant(P0.05).Conclusion:The Caspase-3 protein expression of model rats hippocampus was significantly higher than that of control;Caspase-3 protein expression in Ningshen Wendan decoction group was significantly reduced.It indicates that Ningshen Wendan decoction can control Caspase-3 expression of hippocampal neuronal apoptosis in schizophrenia model rats,which means inhibiting Caspase-3 expression significantly and inhibiting neuronal apoptosis,but the effect of Ningshen Wendan decoction is moderate compared with Chlorpromazine.
