裸鼠正位移植舌癌模型的建立 被引量:2

Establishment of an orthotropic model of human tongue squamous cell carcinoma in nude mice

摘要 目的将舌癌细胞株正位植入裸鼠舌体构建舌癌动物模型,通过模拟舌体内环境,观察肿瘤细胞的生物学行为及淋巴结转移情况。方法将105/0.05 mL Tca83细胞悬液正位移植于裸鼠左侧舌缘,隔日观察记录裸鼠体质量、舌体成瘤情况、肿瘤的长短径;待裸鼠自然死亡或体质量下降至原体质量的25%时,取舌部肿物及颌下淋巴结进行HE及免疫组化检测。结果实验裸鼠在接种后第6~13天相继出现舌部肿瘤,成瘤率为92.11%,通过HE染色及免疫组化染色检测的淋巴结转移阳性率为42.86%。结论成功建立舌癌模型,为研究在体内环境中舌癌的变化发展规律提供了重要的实验手段。通过Tca83所建立的人舌癌动物模型,虽不是最理想的淋巴结转移模型,但因其转移率接近临床统计数据,较好地模拟了人舌癌淋巴结转移情况,为进一步建立高转移率的舌癌模型打下了基础。 Objective To establish an orthotropic model of human tongue squamous cell carcinoma in nude mouse, simulate human body environment and observe the tumor cells biological behavior and condition of lymph node metastasis in vivo. Methods The Tca83 cells were injected into the left margin of tongue in nude mouse. Every location was injected with 105/0.05 mL. The weight, the condition of tumor growth, the length and width of tumor were checked and recorded every two days. Mice were scarified when they died or their weight was less than 25% of primary data. The tongue, lymph nodes were stained with hematoxylin and eosin for microscopic observation, and evaluated for histopathology. Results Tumor was formed on the 6th-13th day after being injected tumor cells. Tumor formation rate was 92.11% , the results of HE staining and immunohisochemistry showed cervical metastasis rate was 42.86%. Conclusions The establishment of an orthotropic model of human tongue squamous cell carcinoma in nude mouse was successful. It supplied an important ex- periment medium in studying tongue cancer development. Although the injected Tca83 cell in nude mouse didn't have an i- deal rate of lymph node metastasis, the result was more close to clinic characteristic and imitated the process of development of human head and neck carcinom The study provided useful information in establishing tongue cancer model with high rate of lymph node metastasis.
