DS对胃癌细胞整合素β1表达及腹腔种植转移的影响 被引量:6

Effects of dextran sulfate on expression of integrin β1 and peritoneal metastasis of human gastric cancer cells in abdominal cavity

摘要 目的探讨硫酸右旋糖苷(dextran sulfate,DS)对胃癌细胞腹腔种植转移的影响。方法采用细胞培养,建立裸鼠动物模型。剖腹并行HE染色观察胃癌细胞腹腔种植转移情况,采用RT-PCR和免疫组化SP法检测DS对黏附分子整合素β1 mR-NA和蛋白的影响。结果剖腹观察瘤节数经HE染色证实,DS实验组腹腔种植转移的瘤节数明显少于对照组(P<0.05)。RT-PCR技术和免疫组化检测结果显示,DS实验组大网膜整合素β1 mRNA和蛋白表达与对照组相比均明显降低,P均=0.000。结论 DS抑制胃癌细胞的腹腔种植转移,且伴有整合素β1表达下降。DS可能是通过下调整合素β1 mRNA影响其蛋白的表达进而抑制肿瘤的种植转移,黏附分子整合素β1的mRNA和蛋白表达下降可能是DS抑制胃癌细胞种植转移的机制之一。 Purpose To investigate the effect of dextran sulfate (DS) on the plantation metastasis of gastric cancer cells in the abdomi- nal cavity. Methods An athymic nude mouse tumor model with gastric cancer cells was established by cell culture technology. By gross exploration and HE staining were carried out to observe the implanted metastasis of gastric cancer cells in the abdominal cavity, and the expression of adhesion molecule integrin β1 mRNA and protein was detected using the reverse transeription-polymerase chain reaction (RT-PCR) and immunohistochemical analysis after DS treatment. Results The number of tumor nodules were grossly ob- served, and confirmed by HE staining. The number of intraperitoneal metastatic nodules in DS experimental group was significantly less than that of control group (P 〈0. 05). By RT-PCR and immunohistochemical analysis, the integrin β1 mRNA and protein in DS ex- perimental group were significantly less than those of control group in omentum ( both P = 0. 000). Conclusion DS inhibits the im- planting metastasis of gastric cancer cells in the abdominal cavity, and one of the underlying mechanisms may be associated with de- creased expression of integrin β1.
