摘要
目的:研究牛蒡提取物对刀豆蛋白A(ConA)诱导肝损伤小鼠的保护作用并探讨其可能的作用机制。方法:小鼠尾静脉注射ConA建立肝损伤模型。计算小鼠肝脏、脾脏和胸腺指数,测定血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)和肝组织中肿瘤坏死因子(TNF-α)和一氧化氮(NO)的含量。观察肝组织病理学变化。结果:牛蒡皮乙醇提取物(PA)显著改善肝损伤小鼠的肝脏和胸腺指数(p<0.05);降低血清中ALT、AST活性和肝组织匀浆中TNF-α、NO的含量(p<0.01);肝脏病变明显减轻。去皮牛蒡根水提物(MA)未表现出护肝作用。结论:牛蒡皮乙醇提取物(PA)对ConA诱导肝损伤小鼠具有保护作用,其作用机制可能与抑制T淋巴细胞的活化和相关炎症因子的释放相关。
Purpose:To investigate the protective effects of burdock concentrate on concanavalin A(ConA)-induced liver injury in mice.Methods:Mice liver injury model was induced by ConA vena caudalis injection.Liver,thymus and spleen index, contents of ALT, AST in serum and TNF- a, NO in liver were measured. Liver histopathological changes were examined accordingly. Results. Compared with modal group, the liver index was decreased and thymus index was increased in PA( peel of burdock extrated by ethanol) group( p 〈0.05).The contents of ALT,AST in serum, and TNF- α, NO in liver homogenate were significantly decreased ( p 〈 0.01 ) .A significant improvement was also observed in pathohistology examination.MW( peeled root extrated by water)had no effect on biochemical indicators and liver histological morphology in mice.Conclusion: PA has protective effect on ConA-induced liver injury in mice. This maybe implementated by inhibiting T lymphocyte activity and decreasing the release of inflammatory factors.
