摘要
目的观察京尼平交联冻存人羊膜对大鼠坐骨神经损伤修复及预防粘连的治疗作用。方法将18只SD大鼠随机分为对照组和京尼平交联冻存人羊膜组(实验组),每组9只大鼠,所有大鼠切除右侧坐骨神经3 mm并行神经外膜缝合。对照组神经缝合处不做处理,实验组神经缝合处用京尼平交联直接快速液氮冻存的人羊膜包裹。术后每日观察大鼠右后足溃疡及愈合情况,术后第10周对神经组织行粘连程度评分及电生理、组织病理学检测。结果实验组术后右后足溃疡愈合时间短于对照组(P<0.05);实验组神经吻合处与周围组织粘连程度小于对照组(P<0.05);实验组吻合处远端坐骨神经再生轴索数大于对照组(P<0.05);实验组比目鱼肌神经-肌肉电潜伏期较对照组短(P<0.05),波幅值较对照组高(P<0.05)。结论经京尼平交联的冻存人羊膜具有促进损伤坐骨神经修复和预防神经粘连的作用。
Objective To observe the effects of genipin-crosslinked cryopreserved human amnion on promoting nerve repair and preventing nerve adhesion in rats with sciatic nerve injury. Methods Eighteen adult rats were randomly divided into 2 groups (n = 9 in each group). Three millimeter dextral sciatic nerve was cut off in all the animals, and the outer membrane of the stumps were sutured. The sciatic nerve was encapsulated by genipin-crosslinked cryopreserved hu- man amnion in the experimental group and not encapsulated in the control group. After surgery, the state of the ulcers and healing status was observed everyday. At 10 weeks postoperatively, the evaluation of adhesion degree, electrophysiolo- gy and histological detections were performed. Results Ulcer healing situation:The healing time in the experiment group were significantly less than that in the control group (P0. 05). The adhesion score of experiment group were less than that of the control group (P0. 05). The fiber hyperplasia was significantly lower in the experiment group than that in the control group. The number of regenerated nerve axons was significantly greater in the experiment group than that in the control group (P0. 05). The peak amplitude of the compound muscle action potential (CMAP) was higher and the CMAP latency was shorter in the experiment group (P0. 05). Conclusion Genipin-crosslinked cryopreserved human amnion can promoting nerve repair and preventing nerve adhesion.
