体内电穿孔技术在DNA疫苗及基因治疗领域的应用进展 被引量:5

摘要 自电穿孔技术诞生以来,其介导的"裸"DNA投递技术已经广泛应用于体外细胞的转染。继而许多研究表明,体内电穿孔技术可以显著提高DNA疫苗或治疗性DNA质粒在肌肉、皮肤、肝、肿瘤及其他不同靶组织的表达水平,显示了良好的临床应用前景。目前,体内电穿孔技术已成功应用于多种实验动物模型,甚至在临床试验中也展示了良好的效果。本文就体内电穿孔技术在DNA疫苗及基因治疗领域的应用进展进行了简要综述。 Since its discovery,electroporation(EP) has been widely used as a mean of introducing "naked" DNA into cells in vitro.And emerging research shows that in vivo EP could significantly enhance the expression of DNA vaccines and therapeutic DNA plasmids in muscle,skin,liver,tumor or other target tissues,suggesting that EP might have potential in clinical application.More recently,in vivo EP has been successfully applied on many animal models,as well as in clinical trial.Here,we briefly review recent advancement in application of in vivo EP in DNA vaccines and gene therapy.
