当归补血汤对大鼠局灶性脑缺血再灌注血-脑屏障和大脑皮质神经元的影响 被引量:11

The effect of Danggui Buxue Decoction on the blood-brain barrier and the cerebral cortical neurons of rats after focal cerebral ischemia reperfusion treatment

摘要 目的:探讨当归补血汤对大鼠局灶性脑缺血再灌注后大脑皮质神经元的治疗作用。方法:将90只雄性SD大鼠随机分为正常对照组、模型组、当归补血汤(高、中、低剂量)组。观察各组大鼠行为学缺陷、从股静脉注入伊文斯蓝溶液观察血-脑屏障通透性变化、用Nissl染色法观察大脑皮质神经元的形态和数量。结果:与模型组相比较,当归补血汤治疗组各剂量均能改善大鼠神经功能(P<0.05)、减少脑组织中伊文斯蓝含量(P<0.05)、增加大脑皮质神经元数量(P<0.05),并以中剂量组效果最好。结论:当归补血汤对缺血再灌注引发的大脑皮质神经元死亡有保护作用,这可能与其降低血-脑屏障通透性有关。 Objective: To explore the therapeutic effect of Danggui Buxue Decoction on the cerebral cortical neuron of rats after the focal cerebral ischemia reperfusion. Methods:90 male SD rats were randomly divided into normal control group, model group and Danggui Buxue Decoction (high, middle and low dose) groups. The differences among groups of the animal behavioral defect, the blood-brain barrier permeability after injection of Evans blue solution through the femoral vein, and the morphology and quantity of cerebral cortical neurons through Nissl staining method were observed. Result: Compared with the model group, all the Danggui Buxue Decoction (high, middle and low dose) groups improved the rats' neurologic function (P 〈 0.05 ), reduced the content of Evans Blue in the brain tissue (P 〈 0.05 ) , and increased the amount of cerebral cortical neuron (P 〈0.05). Among all three treatment (high, middle and low dose) groups, the middle dose group showed the best effect. Conclusion: Danggui Buxue Decoction have therapeutic effect on the loss of cerebral cortical neurons induced by ischemia reperfusion. The underlying mechanisms may be relevant to the fact that Danggui Buxue Decoction can reduce the blood-brain barrier permeability.
