摘要
目的:建立测定异黑成熟颗粒中2种有效成分田蓟苷和甘草酸含量的方法。方法:采用高效液相色谱法,Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,乙腈-0.5%甲酸(27∶73)为流动相,检测波长324 nm,柱温35℃,流速1.0 mL.min-1。Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,甲醇-0.2 mol.L-1酸铵-冰醋酸(67∶33∶1)为流动相,检测波长250 nm,柱温30℃,流速1.0 mL.min-1。结果:田蓟苷和甘草酸分别在1.14~114μg(r=0.999 9)和20.6~206μg(r=0.999 7)线性关系良好;平均回收率分别为98.26%(RSD 2.29%),99.39%(RSD 1.86%)。结论:该方法简便可行,重复性好,可用于异黑成熟颗粒中田蓟苷和甘草酸含量测定。
Objective: To establish the method for the content determination of tilianin and glycyrrhizic acid in Abnormal Savda Munziq granules.Method: HPLC was adopted to determine the content of tilianin and glycyrrhizic acid.The column for Tilianin is Diamonsil C18(4.6 mm ×250 mm,5 μm).The mobile phase was consisted of acetonitrile-0.5% formic acid(27 ∶ 73) with the detection wavelength at 324 nm.The column temperature was set at 35 ℃ and the flow rate was set at 1.0 mL.min-1.The column for glycyrrhizic acid was Diamonsil C18(4.6 mm ×250 mm,5 μm).The mobile phase was consisted of methanol-0.2 mol.L-1 ammonium acetate-glacial acetic acid(67∶ 33∶ 1) with the detection wavelength at 250 nm.The column temperature was kept at 30 ℃ and the flow rate was set at 1.0 mL.min-1.Result: There was good linearity between the peak area and concentration within the range of 1.14-114 μg(r = 0.999 9) for tilianin and 20.6-206 μg(r = 0.999 7) for glycyrrhizic acid respectively.Conclusion: The method is simple,accurate and specific for quality control of Abnormal Savda Munziq granules.
