广东沿海斜带髭鲷群体遗传多样性的初步分析 被引量:1

Preliminary Analysis of Genetic Diversity in Hapalogenys Nitens Stocks in Coastal Waters of Guangdong Province

摘要 目的:运用扩增片段长度多态性(AFLP)技术研究广东省沿海斜带髭鲷野生群体的遗传多样性,并和斜带髭鲷养殖群体做了遗传差异比较。方法:选用9组选择性引物扩增得到353个位点,其中野生群体多态位点175个,多态位点比例为49.58%;人工养殖群体多态位点268个,多态位点比例占75.92%,有7组引物扩增得到的Nei遗传多样性指数和Shannon信息指数斜带髭鲷人工养殖群体比野生群体高;结果:斜带髭鲷野生与人工养殖群体遗传相似系数大,Nei相似系数为0.97,各组引物扩增位点差异比例为0.24。结论:基于NTSYSpc算法构建了实验个体的系统发生树,发现斜带髭鲷野生与人工养殖群体遗传多样性水平低,群体遗传差异不大,由此可初步推断斜带髭鲷野生与人工养殖群体的没有明显遗传分化。 Objective:This paper studies the genetic diversity in wild Hapalogenys nitens stock in coastal waters of Guangdong province in China by means of Amplified Fragment Length Polymorphism (AFLP) technique and then compares the genetic differences between wild and cultured stocks. Method :353 loci were obtained from 9 groups of selective primer amplification of DNA. Among these loci, there are 175 polymorphic loci in wild Hapalogenys nitens stocks at the proportion of 49. 58% and 268 polymorphic loci in cultured stocks at the pro- portion of 75.29%. Nei' s gene diversity index and Shannon' s information index, obtained from 7 groups of primer amplification of cul- tured Hapalogenys nitens stocks, are higher than those of wild stocks. Result: The genetic similarity coefficient of wild and cultured Hapal- ogenys nitens stocks is high, with Nei' s similarity coefficient being 0. 97. The proportion of differences between primer amplification loci is 0. 24. Conclusion:The phylogenetic tree of experimental individuals based on NTSYSpc algorithm indicates a low level of genetic diversity and insignificant genetic differences between wild and cultured Hapalogenys nitens stocks, which can lead to the preliminary inference that there is no distinct genetic differentiation among wild and cultured Hapalogenys nitens stocks.
