摘要
目的对比色法测定乳与乳制品中L(-)-羟脯氨酸的方法和样品处理过程进行探讨。方法采用12.0 mol/L盐酸110(±1)℃恒温消解、过滤后定容、调节pH后按GB/T9695.23-2008加氯胺T氧化、加显色剂、60℃水浴后比色测定,并绘制标准曲线比较定量。结果该法线性范围L(-)-羟脯氨酸在1.0~10.0μg,标准曲线的线性良好,r=0.999 5,方法灵敏度为16.2μg/吸光度,检出限48.8μg/g,回收率在80.5%~95.2%,相对标准偏差在2.15%~4.18%。结论比色法测定羟脯氨酸的准确度和精密度良好,能有效地测定乳及乳制品中L(-)-羟脯氨酸含量。
Objective To discuss the determination of the L ( - ) - hydroxyproline in milk and dairy products and optimized sample processing. Methods Hydrochloric acid of 12. 0mol/L was added by chloramine T oxida- tion and reagent, and underwent 60℃ water bath after 110 + 1 ℃ constant temperature dissolving, filtration volume, adjustment of pH and then determined with colorimetric method according to GB/T9695.23 - 2008, and then the standard curve and quantitative was drew. Results The linear correlation coefficient of standard curve of hydroxyproline was 0. 9995, the detection limit was 48.81xg/g, the sensitivity of the method was 16. 21μg/absorbance, the recovery rate of standard addition was 80. 5% - 95.2%, and the relative standard de- viation was 1.49% -4. 18%. Conclusion With good accuracy and precision, the colorimetrie method can be able to effectively determine the L ( - ) -hydroxyproline content in milk and dairy products.
