摘要
目的 建立抗狂犬病毒 (RV)核蛋白 (NP)荧光标记抗体的制备方法并进行初步鉴定。方法 采用两种方法制备该试剂 :1 从RV感染的细胞中提纯RV核糖核蛋白 (RNP)作抗原 ,免疫动物然后制备荧光标记抗体 ;2 直接用本室原制备的 4株抗RV NP的单克隆抗体混合进行标记。结果 制备的试剂有较高的敏感性和特异性 ,经与法国巴斯德诊断用品公司的相应产品进行比较 ,所得结果的符合率为 10 0 %。特别是用后一种方法制备的试剂优点更多。结论 该制品可替代进口产品 。
Aim\ To establish the methods of preparation of the fluorescein-labeled antibodies to rabies virus(RV) nucleoprotein(NP) and to carry out the preliminary identification.Methods\ Two methods were used to prepare the reagents. 1.The virus ribonucleoprotein(RNP) purified from RV-infected cells was used as antigen and then the fluorescein-labeled antibodies were prepared. 2.The four kinds of monoclonal antibodies to the NP were mixed and labeld with fluorescein. Results\ The reagents prepared have high sencitifity and specificity. The comparison with the reagent produced by Diagnostic Pasteur Inc.(France) showed that these reagents brought identical results.The reagent made by the second method has more advantages. Conclusion\ These reagents could replace the imported product and they could be used in the rapid detection of rabies virus
