摘要
目的研究釉基质蛋白(emdogain,EMD)对体外培养的人牙周膜细胞群(human periodontal ligament cellpopulations,hPDLPs)在牙骨质片上附着、增殖的影响,为EMD联合hPDLPs应用于牙周组织缺损修复提供实验依据。方法采用组织块法分离培养hPDLPs,制备人牙骨质片,实验组用含100 mg/mL EMD的培养液包被处理牙骨质片,对照组用不含EMD的培养液处理。分别于细胞接种牙骨质片上16 h(观察附着情况)和72 h(观察增殖情况),用噻唑蓝比色法检测牙骨质片上的细胞数;同时,用扫描电镜观察牙骨质片上的细胞数。结果噻唑蓝比色法检测,实验组细胞附着数(t=-3.318,P=0.008)、增殖数(t=-6.499,P<0.001)明显多于对照组,两组之间差异有统计学意义。扫描电镜观察计数,实验组hPDLPs附着数(t=8.001,P<0.001)、增殖数(t=13.046,P<0.001)较对照组多,差异有统计学意义。结论 EMD可促进hPDLPs在牙骨质表面的附着和增殖,提示EMD和hPDLPs可联合应用修复牙周组织缺损。
ObjectiveTo observe the effects of enamel matrix protein(EMB) on the attachment and proliferation of human periodontal ligament cell populations(hPDLPs) to root cementum chips in vitro.MethodsCementum chips obtained from the tooth roots were divided into 2 groups.One was experiment group conditioned with EMD,and the other was control group conditioned without EMD.hPDLPs attachment and proliferation were quantified by MTT assay and scanning election microscope at 16 or 72 h.ResultsMTT assay indicated that the number of adhered(t=-3.318,P=0.008) and proliferated(t=-6.499,P0.001) cells on the experiment cementum chips was significantly greater than that on the control.The scanning electron microscope assay indicated that the number of the hPDLPs on the expriment cementum chips was significantly greater than that on the control.ConclusionEMD could promote the attachment and proliferation of hPDLPs on the root cementum chips.It would be helpful for the reconstruction of periodontal tissue defects with EMD combined with hPDLPs.
