摘要
目的探讨2,3-吲哚醌对环磷酰胺诱导的大鼠少弱精症的作用。方法模型组与低中高治疗组连续腹腔注射环磷酰胺(30 mg/kgd)5天,建立大鼠少弱精模型后,正常对照组每天生理盐水灌胃,模型组每天1.25%西黄耆胶水溶液灌胃,低、中、高剂量组分别给予2,3-吲哚醌30 mg/kg、150 mg/kg、300 mg/kg,持续35天。普通光学显微镜观察附睾精子数目、精子活力;放射免疫法检测血清中睾酮(T)、促黄体激素浓度(LH)、促卵泡激素(FSH)含量;HE染色观察生精小管层数;测定睾丸、胸腺、肝、脾重量。结果各剂量治疗组与模型组比较,精子数目增加(P<0.05),a级、a+b级精子比率上升(P<0.05),血清促卵泡刺激素(FSH)下降(P<0.05),生精小管层数增加(P<0.05)。结论 2,3-吲哚醌治疗对环磷酰胺诱导的大鼠少弱精症具有改善作用。
Objective:To study the effect of 2,3-indolinedione(ISA) in the oligozoospermia induced by cyclophosphamide(CP) in rat.Methods:The rat in model group,low-dose,mid-dose and high-dose treatment group were given CP(30 mg/kg·d) through intraperitoneal injection for 5 days,and then give control group normal saline,model group tragacanth(1.25%) and low-dose ISA(30mg/kg),mid-dose ISA(150mg/kg) and high-dose treatment group ISA(300mg/kg) by intragastric administration for 35 days.Measure the sperm mobility rate,sperm counts,serum testosterone(T),follicle-stimulating hormone(FSH),luteinizing hormone(LH) levels,seminiferous tubule layers and the mass of testis,thymus,liver and spleen.Results:Compared to Model group,three treatment groups showed obvious increase of the sperm counts(P0.05),sperm mobility(P0.05) rate and seminiferous tubule layers(P0.05),and decrease of the concentrations of follicle-stimulating hormone(FSH) levels(P0.05) in serum.The mass of testis,thymus,liver and spleen showed no obviously changes.Conclusion:2,3-indolinedione could improve the oligozoospermia induced by cyclophosphamide(CP).
