摘要
目的 探讨小鼠脑缺氧缺血性脑病(HIE)脑组织中水通道蛋白-9(AQP-9)的表达及意义.方法 低氧浓度环境下制作小鼠HIE模型,逆转录-聚合酶链反应(RT-PCR)方法检测AQP-9mRNA的表达水平,免疫组织化学法检测AQP-9蛋白的表达量.结果 模型组各时间点AQP-9mRNA(灰度值最高值1.8223±0.1468)及AQP-9蛋白表达水平[最高值(26.745±2.875)%]均高于对照组,同时AQP-9蛋白与AQP-9 mRNA表达量呈正相关(r=0.811,P <0.01).结论 AQP-9可能参与了小鼠HIE的发生和发展,并起关键作用.
Objective To explore the expression and significance of aquaporin 9 (AQP-9) in the brain tissues of mice with hypoxic-ischemic encephalopathy (HIE).Methods The mouse HIE models were prepared under hypoxia (37,8% oxygen concentration).Immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR) were adopted to detect the expression of AQP-9 protein and mRNA respectively.Results The expression of AQP-9 protein [peak level:(26.745 ± 2.875) %] and mRNA (peak value:1.8223 ± 0.1468) in model group at each every time point was higher than in control group.Simultaneously,there was a positive correlation between AQP-9 protein and mRNA expression (r =0.811,P < 0.05).Conclusion AQP-9 might play roles in formation and development of HIE of mice.
