摘要
目的:研究香青兰总黄酮(dracocephalum total flavones,TFD)对动脉粥样硬化(atherosclerosis,AS)大鼠的作用及其机制。方法:随机将60只雄性Wistar大鼠分为6组,即空白组、模型对照组、TFD高、中、低剂量组(60,30,15 mg·kg-1·d-1)及辛伐他汀组(2.5 mg·kg-1·d-1)。采用高脂饲料喂饲和灌胃维生素D3建立大鼠AS模型。12周后处死大鼠,检测血清血脂及炎症因子水平,采用HE染色及Masson染色观察主动脉病理学变化。结果:与模型组比较,给药组明显降低总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)及炎症因子白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和高敏C反应蛋白(hs-CRP)的水平,明显升高高密度脂蛋白胆固醇(HDL-C)的水平(P<0.01,P<0.05);HE及Masson染色观察TFD给药组表现主动脉粥样硬化组织损伤明显减轻。结论:TFD具有减缓大鼠动脉粥样硬化形成的作用,其作用机制可能与降低血脂水平及抗炎反应有关。
OBJECTIVE To investigate the effect and mechanism of Dracocephalum total flavones (Dracocephalum total fla vones, TFD) on experimental atherosclerosis in rats. METHODS Sixty male Wistar rats were randomly divided into six groups as following: normal control group, model control group, TFD high, medium, low dose group (60, 30, 15 mg'kg-1 d-1 ) and simvastatin group (2. 5 mg.kg . d-a ). Atherosclerotic rat model was established by feeding high-fat diet and treated with VitD3 by ig. After treated 12 weeks, the rats were sacrificed, and then the levels of serum lipid and inflammatory factors were detected. The aortic pathologic changes were measured with HE and Masson staining. RESULTS Compared with model group, the levels of TC, TG, LDL-C and inflammatory factors IL-I, IL-6, TNF-a, hs-CRP were decreased significantly; the level of HDL-C was increased significantly of administration group. The atherosclerosis lesion was significantly reduced in HE and Masson staining of TFD group. CONCLUSION The TFD can inhibit atherosclerosis formation which involved in decrea- sing the levels of serum lipid and anti-inflammatory factors.
