摘要
目的:建立测定茵栀黄口服液中绿原酸含量的方法。方法:采用高效液相色谱法,样品经中空纤维离心超滤法处理后进行测定。色谱柱为Sapphire C18柱,流动相为乙腈-0.4%磷酸溶液(15:85,v/v),检测波长为327nm,流速为1.0ml/min,进样量为20μl,柱温为25℃。结果:绿原酸检测质量浓度在7.5-120μg/ml范围内与峰面积积分值呈良好的线性关系(r=0.9999);精密度、稳定性、重复性试验的RSD≤1.6%;平均加样回收率为98.5%,RSD=1.5%(n=6)。结论:该方法操作简便、结果准确、经济、环保,有利于更加客观、全面地评价及控制茵栀黄口服液的质量。
OBJECTIVE: To establish a method for the content determination of chlorogenic acid in Yinzhihuang oral liquid. METHODS: A hollow fiber based centrifuge ultrafitration pretreatment procedure combined with HPLC was applied to purify solution samples. The separation was carried out on a Sapphire C18 column with acetonitrile-0.4% phosphoric acid solution (15 : 85, V/V) as mobile phase at flow rate of 1.0 ml/min. The detection wavelength was set at 327 nm, the sample size was 20 μl, and the column temperature was set at 25 ℃. RESULTS: The linear range for chlorogenic acid was 7.5-120 μg/mL (r=0.999 9) with an average recovery of 98.5% (RSD=-1.5%, n=6). RSDs of precision, stability and reproducibility tests were all lower than 1.6%. CONCLUSIONS: The method is simple, accurate, economical and environmental, and contributes to objective and comprehensive quality evaluation and control of Yinzhihuang oral liquid.
