甘薯品种F_(3282)凝集素的提取及性质研究 被引量:1

摘要 甘薯品种 F32 82 的浸取液 ,经硫酸铵分级 ,甲壳素层析后 ,再经分子筛过滤 ,获得在 PAGE上呈现单一蛋白质带的甘薯凝集素 ( Ipomoea Batatas L ectin,IBL)。IBL对鸽、兔、人血等红细胞均有凝集作用 ,其中对鸽血的凝集活性最高 ,最低凝集浓度为 0 .0 5μg/ ml。IBL除凝集红细胞外 ,还可凝集小鼠脾细胞和肿瘤细胞 ;其对热相当稳定 ,90℃加热 10 min,仍有部分活性 ;IBL对酸处理较碱处理稳定 ,但可被阿拉伯糖或乳糖等解除凝集活性。IBL的中性糖含量为 5.4 %。 The extraction solution of the Ipomoea batatas F 3282 was graded by ammonium sulphate,chromatographed by the chitin and flitered by molecular sieve.We obtained the IBL(Ipomoea Batatas Lectin)which showed a single of protein on the PAGE.The IBL has agglutination on red blood cells of pigeons,rabbits and human beings.It has the strongest agglutinate activity on pigeonsblood.Its lowest agglutinate concentration is 0.05 μg/ml.Besides red cells,the IBL also can agglutinate the spleen cells of the rats and cancer cells.It remains stable in the heat,so it still has partial activity after being heated 10 minutes under 90 ℃.Being treated with acid,the IBL showed greater stability than when it was treated with alkali.Its agglutination activity was inhibited by arabinose and lactose.The IBL has 5.4% neutral carbohydrates.
