Optimization of fermentation condition for immobilized Sorangium cellulosum

摘要 以多孔陶瓷为载体吸附法固定纤维堆囊菌发酵制备埃博霉素,应用单因素试验和正交设计对固定化纤维堆囊菌发酵制备埃博霉素的发酵条件进行研究和优化。结果表明,用300mL三角瓶进行固定化发酵试验时,其最佳发酵条件为:固液比3:50,温度30℃,转速220 r/min,接种量10%,装液量45 mL,初始pH7.5,发酵时间为8 d。在此条件下埃博霉素产量达40.2 mg/L,与优化条件前相比提高渐近10 mg/L,与游离相比提高了17.5 mg/L。 The Sorangium cellulosum was immobolized on porous ceramics by adsorption, and used to preparation epothilone. Fermentation condition of immobilized Sorangium cellulosum was optimized by single factor design and orthogonal design. The results as follows, the testing of immobilized fermentation with 300 mL flask, the optimal fermentation condition is: solid-liquid ratio 3:50, temperature 30 ℃, shaking speed 220 r/min, inoculum 10%, liquid volum 45 mL, initial pH7.5 and fermentation time 8 d. Under these condition, the epothilone production reached 40.2 mg/L, increased by 10 mg/L than before, and 17.5 mg/ L than floating fermentation.
