摘要
利用PCR-SSCP技术,分析莆田黑猪心脏型脂肪酸结合蛋白(H-FABP)基因的多态性.结果表明:在H-FABP基因5'-调控区检测到两个多态性位点(G7T和C11A),由两个等位基因控制,定义为A和B,基因频率分别为0.740和0.260;在外显子2处存在3个多态性位点(A2C、T62C和T65C),由两个等位基因控制,定义为C和D,基因频率分别为0.164和0.836.群体遗传分析表明,H-FABP基因5'-调控区属中度多态(0.25<PIC<0.5),外显子2区属低度多态(PIC<0.25).χ2适合性检验表明,两位点均处于Hardy-Weinberg平衡状态(P>0.05).将莆田黑猪H-FABP基因外显子2的核苷酸序列与GenBank中野猪核苷酸序列进行同源性比对,同源性为100%,而与牦牛、驴、人、大鼠、鸡和斑马鱼的同源性分别为94.0%、92.4%、87.8%、82.4%、77.9%和67.2%.
Polymorphisms of heart fatty acid-binding protein (H-FABP) gene in Putian Black pigs were detected by PCR-SSCP method. The results showed that two SNPs ( G7T and CllA) were detected in 5' - regnlatory region, and two alleles, designated as A and B, were found. The allele frequencies were 0.740 and 0.260 respectively. Three SNPs (A2C,T62C and T65C) were detec- ted in exord, and two alleles, designated as C and D, were found. The allele frequencies were 0.164 and 0.836 respectively. Sta- tistical analysis revealed an intermediate PIC in 5' - regulatory region (0.25 〈 PIC 〈 0.5 ) but a low PIC in exon2 ( PIC 〈 0.25 ). A chl-square analysis suggested that the allele were in Hardy-Weinberg equilibrium (P〉0.05). The nucleotide sequences of exon2 in Putian Black pig show 100% homology with the same part of Sus scrofa in GenBank, and with Bos gmnniens, Equus asinus, Homo sapiens, Rawa norregicus, Gallus gallus and Danio redo was 94.0%, 92.4%, 87.8%, 82.4%, 77.9% and 67.2% respectively.
