大鼠脾切除后早期自体脾细胞和脾片联合移植对血小板变化的影响

Experimental observation of platelet changes on combination transplantation of antogeneic splenic cell and slices after rat's splenectomy

目的通过建立脾细胞移植和脾片联合移植动物模型来观察移植早期血小板增高恢复状况。方法 Wistar大鼠48只,随机分为4组,其中A组为对照组,行脾切除术,B组为脾片移植组,C组为脾细胞移植组,D组为脾细胞脾片联合移植组,在建立模型前及模型成功后1周、4周、8周抽血,检测血小板水平并进行比较。结果A组脾切除后1、4、8周较术前血小板计数均升高(P<0.05);B组移植后1、4周血小板计数升高(P<0.05),至8周恢复正常(P>0.05),C组移植后血小板计数1、4、8周血小板计数升高(P<0.05),D组移植后1、4周血小板计数升高(P<0.05),至8周恢复正常(P>0.05),在术后1、4周各组血小板计数无显著差异(P>0.05),而在术后8周B和D组与A和C组有差异(P<0.05)。结论脾切除后脾片及联合移植8周后血小板可恢复至正常,单纯脾细胞移植后早期对血小板恢复无明显影响,联合移植对脾切除脾片移植早期血小板恢复也无明显影响。

Objective To observe Spleen graft early thrombocytosis recovery after splenic slices autotransplantation by establishing animal model on combination trantran - splantation of antogeneic splenic cell and slices. Methods Totally 48 Wistar rats are randomly divided into four groups, A group ： splenectomy. B group ： splenic slices transplantation. C group：spleen cell transplantation. D group：combination transplantation. Before animal model and within 1,4,8 weeks af- ter stablishing animal model, peripheral blood is drewn and detected the platelet levels, at the same time, the results are compared. Results In A group postoperative platelet count became higher than preoperation （ P 〈 0. 05 ） ; In B group platelet count became higher at the postoperative 1 th and 4th week（ P 〈 0. 05 ） , but recovered normally at the postopera- tive 8th week（P 〉 0. 05 ） ; In C group postoperative platelet count became higher（ P 〈 0. 05 ） ; In D group platelet count became higher at the postoperative 1 th and 4th week （P 〈 0. 05 ）, but recovered normally at the postoperative 8th week （P 〉 0. 05 ） ; No statistically difference was detected at the preoperative 1 th and 4th week among all the groups for platelet count（P 〉 0. 05 ）. Statistically difference was detected between the B, D and A, C groups for platelet count at the postop- erative 8th week（ P 〈 0. 05 ）. Conclusion After splenectomy, platelets spleen tablets and joint transplantation after 8 weeks recovered to normal, pure spleen cells after transplantation had no significant effect on platelet recovery. United transplant spleneetomy in the spleen graft the early platelet recovery had no significant advantage.
