摘要 目的探讨二型骨形态发生蛋白(BMP-2)和血管内皮生长因子(VEGF)蛋白在大鼠股骨缺损修复过程中的表达和意义。方法 SD大鼠左侧股骨制备1个5 mm缺损,在手术后1周、2周、4周、8周后对大鼠进行X线照射,然后处死所有动物,收获新生骨组织和缺损两端的组织,做苏木素-伊红(HE)染色以及BMP-2和VEGF的免疫组织化学染色。结果 X线检测和HE染色均表明8周时骨缺损在对照组没有愈合,存在骨不连现象;BMP-2和VEGF在手术后第4周时表达达到高峰,并且在骨缺损断端和周围区域表达较缺损中心多。结论持续添加外源性BMP-2和VEGF因子有助于骨缺损的修复,有重要的临床意义。 Objective To investigate the expression of bone morphogenetic protein - 2 ( BMP - 2) and vascular endothelial growth factor (VEGF) in the repair process of femoral defect in rats. Methods A 5 mm femoral defect was prepared in rat, 1,2, 4, 8 weeks after operation, all animals were received X -ray irradiation, and then were killed to harvest the new bone tissues and bone tissues around defect; HE staining and staining of BMP -2 and VEGF were performed with the samples. Results The bone defect was not repaired according to X - ray and HE staining results in control group at 8 w ; and the expression of BMP - 2 and VEGF reached the peak at week 4, the expression of BMP - 2 and VEGF was higher in surrounding area than that in defect center. Conclusion Our results suggest continuous addition of exogenous BMP -2 and VEGF factors can contribute to bone defect, which has important clinical significance.
