摘要 p53是一个众所周知的肿瘤抑制基因,目前的研究表明,其在控制细胞周期动力学、细胞凋亡和肿瘤发生中起着重要的作用。p53的诱导凋亡作用对抑制肿瘤发生至关重要,其能激活多种凋亡相关基因的表达,但其决定性作用的机制尚不明确。p53上调凋亡调控因子(PUMA)与髓细胞白血病基因1(MCL-1)作为p53的靶分子,在决定细胞是否凋亡的过程中起重要作用。该文对细胞凋亡过程中p53基因的激活及其对下游PUMA、MCL-1作用的研究进行综述。 p53, a well known tumor suppressor gene,plays a major role in the control of cell cycle kinetics,cell death and carcinogenesis. As a tumor suppressor gene, its activation and regulation mechanism is a hot spot. Several stresses lead to p53 activation, which results in various biological outcomes, including cell cycle arrest and apoptosis. The exact decisive role of p53 is unclear. PUMA and MCL-1 are p53 target molecules which play an important role in the process of cell apoptosis. Here is to make a review of the activation of the p53 gene and its downstream PUMA,MCL-1 role.
