脲酶聚丙烯酰胺凝胶电泳活性区带染色方法的建立与应用 被引量:1

摘要 目的 :建立脲酶聚丙烯酰胺凝胶电泳活性区带染色方法用于测定脲酶分子量和纯度鉴定。方法 :建立十二烷基硫酸钠-聚丙烯酰胺凝胶电泳 (SDS -PACE)梯度凝胶体系和电泳条件等 ,根据不同来源脲酶活性染色区带位置 ,鉴定出PAGE蛋白区带中脲酶区带位置 ,进一步测出脲酶的相对分子量。结果 :用新建立的SDS -PAGE脲酶活性区带染色方法 ,应用于不同来源的脲酶相对分子量测定和纯度鉴定 ,结果准确 ,重复性好 ,与凝胶过滤法测定分子量的结果相关性好。结论 :我们建立的SDS -PACE脲酶活性区带染色方法 ,应用效果良好 ,有推广价值。 Objective:Set up polyacrylamide gel electrophresis(PAGE)activity zone staining method for measuring urease molecular weight and determining purity.Methods:Set up SDS-PAGE gel series and electrophresis condition,etc.According to position of different source urease activity staining zone to determine urease zone of PAGE protein zones,get advanced measuring urease molecular weight.Results:[WT5BZ]We made use of new SDS-PAGE urease activity zone staining method to measure different source urease molecular weight and to determine purity.The result indicated that It was acurate and repeatable very well.It was highly correlative to compare with result of gel filtration.Conclusion:Effect of SDS-PAGE urease activity zone staining method is very well.The method is worthy of spreading.
