摘要
交链孢霉是污染粮食、蔬菜、烟草等植物的常见霉菌,该霉菌能产生多种毒性代谢物,交链孢酚单甲醚(Alternariol Monomethyl Ether简称AME)是其中主要毒素之一,本文介绍了将AME重氮化,然后将AME偶氮化合物与牛血清白蛋白(BSA)进行交联制成完全抗原,用AME-BSA完全抗原免疫家兔制备抗AME抗体。并将AME标记上半乳糖苷酶,用标准AME与之竞争,通过ELISA竞争试验,绘出标准曲线图,证明所制备的抗AME抗体能与标准AME进行特异性结合,可用于粮食中AME含量的检测。
Alternaria alternata is a usually contaminative mold of plants, such as grain, vegetable and tobacco, This mold can produce many kinds of toxin metablite. The major one of them is AME, The procedure to prepare the anti-AME antibody is introduced here. AME is a hapten. AME was first diazotized, then a complete antigen (AME-BSA) was prepared by conjugation of diazo-ompound and bovine serum albumin. AME-BSA antigen was injected into rabbits to prepare anti-AME antibody By different coupling reagents and different coupling method AME was labelled B-galactosidase. with competive ELISA the standard curve was established and this showed that prepared anti-AME antibody could couple withAME specially . These experiments demonstrated that prepared anti-AME antibody can be used to detect the quantity of AMNin grains.
