摘要
目的:探讨原发性肝细胞癌(简称肝癌)细胞凋亡相关蛋白bcl-2和bax的表达情况及其意义。方法:采用免疫组化ABC法检测bcl-2和bax蛋白在66例人肝癌组织和24例人正常肝组织,以及3种人肝癌细胞系HCC-9204、SMMC-7721、HHCC和1种人正常肝细胞系QZG的表达情况,并分析肝癌组织的bcl-2和bax蛋白表达阳性率与其病理分级之间的关系。结果:bcl-2蛋白在正常肝组织中表达的阳性率只有4.2 %(1/24),在肝癌组织中的阳性率为25.8 %(17/66),二者具有显著性差异(P<0.05)。bax蛋白在正常肝组织中表达的阳性率为70.8 %(17/24),在肝癌组织中的阳性率为43.9 %(29/66),二者也具有显著性差异(P<0.05)。肝癌组织的bcl-2和bax蛋白表达阳性率与其病理分级之间均未见明显的相关性。在正常肝和肝癌组织中,bcl-2蛋白的阳性率均明显低于bax蛋白(P<0.01,P<0.05)。在正常肝细胞系QZG中未见明显的bcl-2阳性信号,bax蛋白的阳性细胞计数率为61.0%。肝癌细胞系SMMC-7721和HHCC的bcl-2蛋白阳性细胞计数率分别为14.3%和11.8%,bax蛋白阳性细胞计数率分别为58.3%和47.1%。肝癌细胞系HCC-9204中未见明显的bcl-2阳性信号,bax蛋白阳性细胞计数率为43.2%。结论:与正常肝组?
Objective To explore the expression and significance of apoptosis-associated proteins bcl-2 and bax in primary hepatocellular carcinoma HCC. Methods The expression of bcl-2 and bax proteins in 66 cases of human HCC tissues.24 normal liver tissues.3 HCC cell lines HCC-9204.SMMC-7721 and HHCC and a human normal hepatocyte cell line QZG were detected with immunohistochemical ABC method. The relationship between the pathological grades of HCC tissues and their positive rates of expressions of bcl-2 and bax proteins were analysed. Results The positive rate of bcl-2 protein in normal liver tissues was only 4.2% 1/24.while that in HCC tissues was 25.8% 17/66 with significant difference between them P<0.05. The positive rate of bax protein in normal liver tissues was 70.8% 17/24.while that in HCC tissues was 43.9% 29/66 also with signifiicant difference between them P<0.05. No siginificant correlation was observed between the positive rate of bcl-2 or bax priotein expression in HCC tissues and their pathological grades. Both the positive rates of bcl-2 and bax proteins in normal liver tissues were significantly lower than those in HCC tissues P<0.01.P<0.05. There was no detectable bcl-2 positive signal.and the countable rate of bax protein positive cells was 61% in normal hepatocyte cell line QZG. The countable rate of positive bcl-2 protein cells in SMMC-7721 and HHCC were 14.3% and 11.8%.and those of positive bax protein cells were 58.3% and 47.1% in HCC cell line SMMC-7721 and HHCC respectively. There was no detectable positive bcl-2 signal.and the countable rate of positive bax protein cells was 43.2% in HCC cell line HCC-9204. Conclusion Compared to normal liver tissues.there is low-level increase of bcl-2 protein expression and decrease of bax protein expression in HCC tissues. The expression levels of bcl-2 protein are lower than those of bax protein in both normal liver and HCC tissues. The expression level of bcl-2 is very low.and there is a moderate-level bax protein expression in HCC and normal liver hepatocyte cell lines.
