烟粉虱与温室白粉虱羧酸酯酶、谷胱甘肽转移酶和乙酰胆碱酯酶性质的比较研究 被引量:30

摘要 对温室白粉虱 Trialeurodes vaporariorum和烟粉虱 Bemisia tabaci羧酸酯酶(Car E)、乙酰胆碱酯酶 (ACh E)和谷胱甘肽转移酶 (GSTs)的生物化学性质进行了初步研究。比较 Car E活性 ,烟粉虱显著高于温室白粉虱 ,烟粉虱和温室白粉虱 Car E对底物乙酸 -α-萘酯的 Km 分别为 0 .0 73mmol/ L 和 3.4 5 8mmol/ L。烟粉虱 ACh E活性分布高峰在 2~3m OD/ (min·头 )之间 ,而温室白粉虱 ACh E活性分布高峰在 10~ 15 m OD/ (min·头 )之间 ,烟粉虱 ACh E的 Km 值是温室白粉虱 Km 值的 12 .6倍。烟粉虱的 GSTs比活力为2 .2 18OD/ (mg pro·min) ,显著高于温室白粉虱的 GSTs比活力 0 .6 6 3OD/ (mg pro· min) ,差异达极显著水平 ,烟粉虱 GSTs对底物的亲和力比温室白粉虱高。 The characteristics of carboxylesterase, glutathione S transferase and acetylcholinesterase were investigated in Trialeurodes vaporariorum , fed on pumpkin, and Bemisia tabaci , fed on cotton. The comparative studies revealed that the resistance mechanisms of T. vaporariorum may be distinguished with that of B. tabaci in carboxylesterase(CarE), acetylcholinesterase (AChE) and glutathione S transferases(GSTs). The activity of CarE in B. tabaci strain was higher than that in T. vaporariorum while the comparision of Michaelis constants( K m) of CarE between B. tabaci and T. vaporariorum showed that the affinity of the former to substrate was much stronger than that of the latter. The activity of AChE mainly distributed in 2~3 mOD/(min· fly) in B. tabaci on cotton and 10~15 mOD/(min·fly) in T. vaporariorum on pumpkin, respectively. The activity of AChE in B. tabaci on cotton was far lower than that in T. vaporariorum . The comparison of Michaelis constants ( K m) of AChE between T. vaporariorum and B. tabaci indicated that the affinity of B. tabaci on cotton to substrate was much weaker than that of T. vaporariorum strain. The activity of GSTs of B. tabaci was far higher than that of T. vaporariorum and the affinity of the former to substrate was stronger than that of the latter.
