摘要 目的 :通过研究 β 内酰胺抗生素与Superdexpeptide凝胶介质的相互作用 ,为建立和优化 β 内酰胺抗生素中高分子杂质的分离分析方法提供理论参考。方法 :考察 8种 β 内酰胺抗生素在Superdexpeptide凝胶介质中不同色谱条件下的色谱行为 ,并与其在TSKG2 0 0 0SW亲水性多孔硅胶介质中的色谱行为比较。结果 :β 内酰胺抗生素在Superdexpeptide和TSKG2 0 0 0SW介质中的保留行为受到流动相缓冲盐组分、pH值、离子强度的影响。结论 :β 内酰胺抗生素在凝胶介质中受到分子排阻、吸附作用的共同影响 ,不能简单地以保留体积判定 β Objective: To develop a novel method for separation and analysis of high molecular weight impurities and optimize method through studying the interaction between β lactam antibiotics and gel matrix of Superdex peptide. Method: Eight β lactam antibiotics with different structures were studied under different chromatographic conditions in gel matrix of Superdex peptide, and the chromatography behaviour in Superdex peptide was compared with that in TSK G2000SW. Results: The chromatography of β lactam antibiotics in Superdex peptide and TSK G2000SW was influenced by buffer composition, pH value, and ionic strength of mobile phase. Conclusion: Because of the chromatography of β lactams antibiotics in gel matrix was influenced by size exclusion, adsorption and other factors, the molecular weight of β lactam antibiotics impurities could not be identified only by its retention volume.
