摘要
目的:研究氦氖激光对退变颞下颌关节髁突软骨细胞外基质和基质调节因子表达的影响。方法:将40只健康成年雄性新西兰大白兔随机分为正常组、假模型组、模型组和治疗组(n=10),对治疗组兔采用氦氖激光治疗,组内又分为造模后第1、11天2个取材时间点(n=5)。于不同时间点取出颞下颌关节组织,HE染色观察组织学形态,Western Blot方法检测细胞外基质Col-2、PRG-4和基质调节因子MMP-13、TIMP-1、BMP-2的表达。结果:经过氦氖激光治疗后的髁突软骨纤维层轻度松解和部分胶原纤维合成增强。造模后第1天模型组与治疗组MMP-13表达均明显升高,Col-2、PRG-4、TIMP-1、BMP-2表达均明显降低(P<0.05);治疗组第11天与第1天比较,MMP-13表达降低(P<0.05),Col-2、PRG-4、TIMP-1、BMP-2表达升高(P<0.05)。结论:氦氖激光治疗可上调退变颞下颌关节软骨细胞外基质Col-2、PRG-4和基质调节因子TIMP-1、BMP-2蛋白表达,下调MMP-13蛋白表达。
Objective:To observe the effects of He-Ne laser on the expression of extracellular matrix and its regulatory factors in degenerated temporomandibular joint(TMJ)of rabbits.Methods:40New Zealand adult white rabbits were randomly divided into normal group,sham model group,TMJOA model group and laser treatment group(n=10),the rats in treatment group were treated by He-Ne laser irradination at acupoints.The rats in each group were divided into1d and11d groups(n=5).The animals were respectively killed1d and11d after operation and HE staining was used to observe the histomorpholy.The protein expression of extracellular matrix and its regulatory factors were examined by Western blot.Results:After He-Ne laser treatment,the fiber layer of condylar carti- lage was slightly loose,part of the fiber was newly produced.The level of Col-2,PRG-4,TIMP-1,BMP-2was up-regulated and the of MMP-13was down-regulated in the11d treatment group.Conclusion:He-Ne laser irradiation on acupoints may up-regulate the expression of extracellular matrix(Col-2and PRG-4)and its regulatory factors(TIMP-1and BMP-2),down-regulate the expression of MMP-13.
