摘要
目的优选出薄荷脑和冰片β-环糊精(β-CD)的最佳包合工艺饱和水溶液法,以包合物收得率,薄荷脑、冰片包合率为评价指标,通过正交设计实验分别考察β-CD与薄荷脑和冰片的质量比、包合温度、包合时间对包合工艺的影响;通过薄层色谱法和红外光谱法对包合物进行验证;以薄荷脑和龙脑的相对含量为指标,考察其稳定性。结果最佳包合工艺为:β-CD与薄荷脑和冰片的质量比6∶1,包合温度30℃,包合时间1.5 h;薄层色谱法和红外光谱法均表明β-CD包合物的形成;包合物在空气、高温和高湿条件下的稳定性明显高于其物理混合物。结论优选出的包合工艺收得率和包合率较高、工艺稳定、合理可行。
Objective To study the optimum inclusion process for menthol and borneol with β-cyclodextrin(β-CD). Methods The menthol and borneol are encapsulated by saturated aqueous solution method. The yield of inclusion compound, the inclusion rate of menthol and borneol are used as evaluating indicators to investigate the effects of three factors, including the mass ratio of β-CD to menthol and borneol, inclusion temperature and inclusion time by orthogonal design experiments;and the inclusion compound is confirmed by thin-layer chromatography(TLC) and infrared spectroscopy(IR);the relative contents of menthol and borneol are used as indicators to investigate the stability of inclusion compound. Results The optimum inclusion process for menthol and borneol is as follows: the mass ratio of β-CD to menthol and borneol is 6 : 1, the inclusion temperature is 30 °C, and the inclusion time is 1.5 h;both thin-layer chromatography and infrared spectroscopy indicated that menthol and borneol β-CD inclusion compound is formed;the inclusion compound is significantly more stable than the physical mixtures when exposed to air, high temperature and high humidity. Conclusion The optimum inclusion process has good yield and inclusion rate, and the process is stable and reasonable.
