摘要
目的:建立以高效液相色谱法测定皂角刺中槲皮素和刺囊酸的含量。方法:采用VenusilXBPC18(250mm×4.6mm,5μm)色谱柱,流动相为乙腈-0.05%磷酸水,梯度洗脱,检测波长203nm,流速:1mL·min-1,柱温:30℃。结果:槲皮素、刺囊酸分别在0.012~0.24mg·mL-1、0.016~0.32mg·mL-1范围内呈现良好的线性关系,相关系数r分别为0.9999和0.9996,平均加样回收率分别为99.70%(RSD1.56%)、101.4%(RSD2.32%)。结论:该方法简单、快捷、准确,可以同时测定皂角刺中槲皮素和刺囊酸的含量,为中药饮片皂角刺的质量控制提供实验数据。
Objective:To establish a method for the determination of quercetin and echinocystic in Spina Gleditsiae. Methods:A Venusil XBP C18 column (250 mm ×4.6 mm,5 μm)was adopted,and with acetonitrile -0.05% phosphoric acid as the mobile phase for gradient elution.The detection wavelength was set at 360nm.The flow rate was 1.0 mL·min^-1 and the column temperature was 30 ℃ .Results:The calibration curve showed good linearity for quercetin and echinocystic acid in the concentration rage of 0.012-0.24mg·mL^-1 (r=0.9999)and 0.016-0.32 mg·mL^-1(r=0.9996). The average recoveries of quercetin and echinocystic acid were 99.70%(RSD 1.56%)and 101.4%(RSD 2.32%).Conclusion:The method is simple and accurately.It can determine quercetin and echinocystic acid at the same time.The method provide experimentation data for the quality control o f Spina Gleditsiae.
