摘要
目的 观察烧伤大鼠血清对肠上皮细胞膜损害的影响。方法 制作烧伤大鼠模型 ,采集血清 ;肠上皮细胞株IEC 6经烧伤大鼠血清刺激后 ,细胞膜磷脂分析采用高效毛细管电泳法 ;膜流动性测定用荧光偏振法 ;并对膜丙二醛 (MDA)含量和超氧化物歧化酶 (SOD)活性进行测定。结果 IEC 6经烧伤血清作用后 ,早期即出现细胞膜流动性下降 ;膜磷脂含量降低 ,组分改变 ;SOD活性持续降低 ;但MDA含量增加。结论 烧伤后血清中的有害物质可致肠上皮细胞膜磷脂降解、脂质过氧化增强 ,导致膜流动性降低 。
Objective To observe the effects of burned rat serum on the intestinal epithelial cellular membrane damage. Methods The intestinal epithelial cell IEC 6 were stimulated with the serum collected from the 30% TBSA Ⅲ degree model rats. The changes of the membranous phospholipid contents were analyzed by the high performance capillary electrophoresis. The fluidity of the cell membrane was dynamically measured with fluorescence polarization technique.The content of MDA and the level of SOD in the cell membrane were tested respectively by the biochemical kit. Results Membramous fluidity and the phospholipid contents decreased obviously in the early stage in IEC 6 stimulated by the serum of burned rat. Meantime, the level of SOD in the membrane decreased but the content of MDA significantly increased. Conclusion The lipid peroxidation and the degradation of the membranous phospholipid induced by the serum of burned rat might be one of the factors of the decrease of membrane fluidity, and this may be an important mechanism of the damage of membrane structure and function in IEC 6.
