摘要
研究益气活血方剂抑制 HSC合成 型胶原的作用和生物学机制。采用免疫性肝纤维化大鼠模型 ,制备不同造模时间的病理血清、正常血清和益气活血方药物血清培养肝星形细胞 HSC (Hepatic stellate cells) ,激光共聚焦显微镜定量分析 型胶原的表达。结果显示 :1正常大鼠血清培养继代 HSC表达较多的 型胶原。纤维化模型大鼠血清诱导 HSC表达的 型胶原低于正常大鼠 ,益气活血药物血清可抑制纤维化模型血清诱导的 HSC 型胶原的表达 ,P<0 .0 5。 2 3周模型血清培养的 HSC中分别加入 1 0 - 6 、 1 0 - 7和 1 0 - 8ngγ-干扰素表明 ;1 0 - 6 、 1 0 - 7ngγ-干扰素和益气活血方具有相同的抑制 HSC表达 型胶原的作用 ,1 0 - 8ngγ-干扰素抑制作用次之 ,P<0 .0 1。 31 %胎牛血清可使原代 HSC表达低水平 型胶原。结果表明 :益气活血方剂能够抑制 HSC合成 型胶原 ,其作用机制是改变了 HSC的生活状态。
This paper reported that the recipe of YIQI HUOXUE inhibited effect on collagen Ⅰ expression in HSC. HSCs were cultured in conditional media with rat sera. The conditional media consisted of 4 groups. Group1: DMEM contained 10% normal rat serum. Group2: DMEM contained 10% hepatic fibrotic rat serum which modeled for 3 weeks, 5 weeks and 7 weeks respectively. Group3: DMED was added 10% rat serum in the recipe of YIQI HUOXUE into group2. Group4: 10 -6 ng, 10 -7 ng and 10 -8 IFN γ was added to DMEM which contained hepatic fibrotic rat serum at 3 week. Expression of collagen Ⅰ was quantifyed by laser common focus microscope. The results showed that more collagen Ⅰ in group 1 was expressed than that of group2 in subculture HSC. The expression of collagen Ⅰ in group 2 could be inhibited by group 3 in subculture HSC, P <0 05. IFN γ 10 -6 ng and 10 -7 ng of group 4 could inhibit the expression of collagen Ⅰ as group 3 in subculture HSCs. The effect of IFN γ 10 -8 ng was less than that of group 3, P <0 01. In addition, the expression of collagen Ⅰ was observed in primary HSC, the result was similar to that of subculture HSC. It is indicated that the recipe of YIQI HUOXUE can inhibit expression of collagen Ⅰ in HSCs, and the result suggusts that the state of HSC can be reversed by the recipe of YIQI HUOXUE.
