不同孕期人滋养层细胞表皮生长因子受体及其mRNA表达的研究 被引量:4

摘要 目的:了解不同孕期人滋养层细胞表皮生长因子受体(EGFR)及EGFRmRNA的表达,进而探讨其在胎盘分化及发育中的作用。方法:采用免疫组织化学方法测定不同孕期滋养层细胞EGFR的表达,应用逆转录-聚合酶链反应(RT-PCR)技术对EGFRmRNA进行分析。结果:EGFR存在于妊娠各期滋养层细胞的细胞膜、核膜及胞质内,既表达于合体滋养层细胞(ST),也表达于细胞滋养层细胞(CT)。孕早期EGFR的表达强于妊娠中期和晚期。EGFRmRNA在各期滋养层细胞的表达从强到弱依次是孕早期、中期、晚期。结论:EGFR及其mRNA在不同孕期滋养层细胞表达的强度不同,其中孕早期较妊娠中期、晚期表达明显增强。 Objective: To study the expressions of epidermal growth factor recept or(EGFR) and EGFR mRNA in human trophoblast cells of different pregnancy phases ,and to explore the effect of EGFR on cell proliferation and development. Metho ds:The EGFR expression in human trophoblast cells of different pregnancy phases was localized by immunohistochemical methods(ABC),and the related quantity of EGFR mRNA was analyzed by RT-PCR method. Results:EGFR protein was distributed throughout the cell membrane,nuclear membrane and cytoplasm,not only in syncyt iotrophoblast,but also in cytotrophoblast.Expressiong of EGFR in placenta of th e first trimester was significantly richer than that in placenta of the second a nd third trimesters.The related quantity of EGFR mRNA expression in the placenta of the first trimester was significantly stronger than that in placenta of the second and third trimesters. Conclusions:EGFR was expressed in human trophoblas t cell of different pregnancy phase,and it was significantly higher in the firs t trimester than in the second and third trimesters.
