腺苷对未成熟心肌缺血再灌注损伤的保护作用

摘要 目的 研究腺苷对离体未成熟心肌的保护作用。 方法 采用出生 (21±2)d的健康小白兔离体心脏 ,建立离体心脏左心做功模型 ,随机分为加腺苷组 (4℃ST.ThomasⅡ停搏液 +腺苷200μmol/L)和对照组 (4℃ST.ThomasⅡ停搏液 ) ,观察腺苷停搏液对心脏缺血120min再灌注60min后心脏功能、心肌酶、心肌丙二醛含量和超微结构变化的作用。 结果 腺苷组和对照组比较 ,停搏液加入腺苷者心功能恢复良好 (P<0.05) ,冠脉流量提高 (P<0.05) ,心肌酶含量降低 (P<0.05) ,心肌超微结构改变明显减轻。结论 腺苷能提高ST. Objective To determine the protective effect of adenosine enrichment to ST.Thomas′ Hosptial cardioplegic solution(STHCS) Ⅱon ischemic immature myocardium under repertusion injury.Methods Isolated working rabbit hearts[(21±2)days] were subjected to 120 min of hypothermic(4℃) cardioplegic arrest and 60 min of normothermic(37℃) reperfusion.The adenosine group(group B) received STHCS Ⅱ with adenosine (200μmol/L) enrichmentand and the control group (group A) was treated with STHCS Ⅱ.Cardiac fuction,myocardial MDA,coronary flow were measured and electron microscopic examination was studied.Results Recovery of cardiac fuction was significantly better in group B.The myocardial MDA?LDH,CPK and CPK_MB levels in group A were significantly higher than that in group B(P<0.05).The coronary flow increased in group B after reperfusion(P<0.05).Electron microscopic examination show relatively minimal myocadial cell injury in group B as compared with that in group A. Conclusion These results suggest that the myocardial protective effect of STHCS Ⅱon immature hearts can be enhanced by adenosine.
