摘要
目的探讨氧化应激在肌萎缩侧索硬化(ALS)发病中的作用及其与病情评价指标的相关性。方法应用酶联免疫吸附测定(ELISA)法、硫代巴比妥酸法以及黄嘌呤氧化酶法分别测定39例ALS患者和43例正常对照者血清中8-羟基脱氧鸟苷酸(8-OhdG)、丙二醛(MDA)、谷胱甘肽(GSH)的水平;采用量表评价病情严重程度。结果ALS组血清中8-OhdG(195.02±51.47)ng/L和MDA(8.34±1.67)nmol/L明显高于对照组(122.47±56.87)ng/L和(3.47±1.17)nmol/L(P<0.05);ALS组血清中GSH(18.34±2.55)nmol/L明显低于对照组血清中GSH(28.34±3.87)nmol/L(P<0.05)。ALS组血清中8-OhdG、MDA及GSH的水平与病情严重程度有相关性(P<0.05)。结论ALS患者出现了明显的氧化损伤;血清中8-OhdG、MDA及GSH的水平与ALS病情轻重相关。
Objective To explore the effects of oxidative stress in amyotrophic lateral sclerosis(ALS)and its relevance with the disease evaluation index.Methods The level of 8-hydroxy deoxyguansine acid(8-OhdG),malondialdehyde(MDA)and glutathione(GSH)in 39 cases of ALS and 43 normal controls were detected by enzymelinked immunosorbent assay(ELISA),thiobarbituric acid and xanthine oxidase technique.The severity of disease was evaluated by using rating scale.Results The 8-OhdG(195.02±51.47)ng/L and MDA(8.34±1.67)nmol/L in the serum of patients were significantly higher than those in the control group(122.47±56.87)ng/L and(3.47±1.17)nmol/L(P 0.05).The GSH level(18.34±2.55)nmol/L in the serum of patients was significantly lower than that in the control group(28.34±3.87)nmol/L(P 0.01).2There were relevance between the level of 8-OhdG,MDA and GSH and the disease severity in the patients(P 0.05).Conclusion Obvious oxidative damage exists in ALS patients.The levels of 8-OhdG,MAD and GSH are closely related to the severity of ALS patients.
