摘要
目的观察参芍口服液对心肌缺血再灌注损伤大鼠基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶抑制因子-2(TIMP-2)和心肌激酶的影响。方法选取40只SD大鼠,随机平均分为4组,即假手术组(A组)、缺血再灌注组(B组)、参芍口服液组(C组)、瑞舒伐他汀组(D组),每组10只。C组、D组分别于术前3 d给予参芍口服液8ml/d、瑞舒伐他汀10 mg/d灌胃,A组、B组给予生理盐水。采用结扎大鼠冠状动脉左前降支30 min再灌注120 min的方法复制大鼠心肌缺血再灌注模型(假手术组不结扎)。分析各组大鼠心肌HE染色情况、MMP-2和TIMP-2的表达、心肌激酶变化。结果心肌HE染色:A组心肌结构正常,B组心肌细胞破坏最严重,C组心肌细胞破坏程度较B组和D组明显减轻。免疫组化检测:MMP-2蛋白的表达水平A组<C组<D组<B组,各组间相比有统计学意义(P<0.05);TIMP-2蛋白的表达水平A组>C组>D组>B组,各组间相比有统计学意义(P<0.05)。大鼠心肌酶:CK的水平A组有很少量的表达,其余各组均有大量表达(P<0.05);CK-MB、LDH的水平A组<C组<D组<B组,各组间相比有统计学意义(P<0.05)。结论参芍口服液预处理对大鼠心肌缺血再灌注有保护作用,其作用机制可能与降低MMP-2、升高TIMP-2的表达有关。
Objective To observe the effect of Shenshao oral liquid on myocardial ischemia reperfusion injury , matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2)and myocardial kinase of rat model. Methods 40 SD rats were randomly divided into 4 groups, namely sham operation group (A group), ischemia reperfusion group (B group) and Shenshao oral liquid group (C group) and rosuvastatin group (D group), 10 rats in each group.C group and D group were given 3 days of Shenshao oral liquid for 8 ml/d, rosuvastatin 10 mg/d orally before operation , A group, B group received saline .By using the method of the left coronary artery ligation on rats anterior descending artery for 30 min, then reperfusion for 120 min to replicate myocardial ischemia reperfusion rats model ( sham operation without liga-tion) .Analyze rat myocardial HE staining , MMP-2 and TIMP-2 expression and myocardial kinase changes .Results Myocar-dial HE staining:A group myocardial structure was normal , the most serious damage to myocardial cells was in B group , C group ’ s myocardial cell damage was milder than that in group B and D group .Immunohistochemical detection:MMP-2 protein expression level was A group 〈C group 〈D group 〈B group, there was statistical significance ( P 〈0.05);TIMP-2 protein expression level was A group 〉C group 〉D group 〉B group, there was statistical significance among each group ( P 〈0.05).Rat myocardial enzyme: level of CK expression was very small in A group , the other groups were expressed a lot (P 〈0.05); CK-MB, LDH level: A group 〈C group 〈D group 〈B group, there were statistical significance ( P 〈0.05).Conclusion The Shenshao oral liquid preconditioning on myocardial ischemia reperfusion injury in rats has a protec -tive effect, the mechanism may be related to the decrease of MMP-2 and increase the expression of TIMP-2.
