针灸“肺俞”穴对病毒性肺炎小鼠肺指数及相关炎性因子的影响 被引量:33

Effect of Electroacupuncture Stimulation of “Feishu” (BL 13)on Lung Index,Serum and Lung IL-10 and TNF-α Levels in Mice with Viral Pneumonia

摘要 目的:观察针灸"肺俞"穴对病毒性肺炎小鼠肺指数及其肺组织和血清中白介素(IL)-10、肿瘤坏死因子(TNF)-α水平的影响,探讨针灸抑制病毒性肺炎的效应机制,为针灸介入小儿病毒性肺炎的防治提供实验依据。方法:健康KM小鼠采用流感病毒亚洲甲型鼠肺适应株滴鼻感染复制病毒性肺炎模型,造模后小鼠分为模型组、西药组、电针组和艾灸组,同时选健康KM小鼠作为正常对照组,每组10只。电针组及艾灸组电针及艾灸小鼠背部双侧"肺俞"穴,每日灸或针刺两次,共治疗1周;西药组腹腔注射炎琥宁,每日1次,治疗1周。计算各组小鼠肺指数,采用固相夹心法酶联免疫吸附实验检测小鼠肺组织及血清中IL-10、TNF-α水平,观察各组肺组织病理变化。结果:模型组肺指数及血清TNF-α、肺组织TNF-α及IL-10水平较正常组明显升高(P<0.01)。西药组、电针组及艾灸组肺指数及血清TNF-α均较模型组降低(P<0.01),电针组较西药组降低更明显(P<0.05)。造模后血清IL-10较空白组变化不明显(P>0.05),各组治疗后血清IL-10较模型组升高(P<0.01)。各治疗组肺组织TNF-α及IL-10均较模型组降低(P<0.05,P<0.01)。正常组小鼠肺脏无充血水肿,模型组肺脏充血水肿明显,西药组、电针组和艾灸组小鼠肺脏轻度充血水肿。结论:针灸"肺俞"穴辅助治疗病毒性肺炎可能是通过抑制或调节相关炎性因子IL-10、TNF-α水平来发挥作用,同时通过进一步抑制TNF-α的过量产生而减轻病毒感染引起的免疫病理损害。 Objective To observe the effect of electroacupuncture (EA) stimulation of "Feishu'(BL13) on lung pathological changes, and levels of pulmonary and serum IL-10 and tumor necrosis factor (TNF)-α in viral pneumonia mice, so as to explore its mechanisms underlying improvement of viral pneumonia. Methods Fifty Kunming mice were randomly divided into normal control, model, medication, EA and moxibustion groups, with 10 mice in each group. The viral pneumonia model was estab- lished by intra-nose drip of influenza A1 virus FM1 (H1N1). Rats of the EA group were treated by EA stimulation of bilateral BL13, those of the moxibustion group treated by ignited moxa-stick, and those of the medication group treated by intraperitoneal injection of Yanhuning injection (10 mg · Kg-1 · d^-1 , solved in 5% glucose solution, 5 mL/time/mouse, once daily for 1 week). Both EA and moxibustion treatments were conducted for 20 min, twice a day for a week. The lung index i-lung weight/body weight × 100% ] were calculated after the treatment. Pathological changes of the lung were observed under microscope after H.E. stai- ning, and serum and pulmonary IL-10, TNF-α levels were detected using solid phase sandwich enzyme-linked immunosorbent assay. Results Following modeling, the average lung index, serum TNF-α, and pulmonary TNF-α and IL-10 levels were signifi- cantly increased in the model group (P〈0.05), while after the treatment, the average lung index and serum TNF-α and pulmonary IL-10 levels in the EA, moxibustion and medication groups were notably down-regulated (P〈0.05, P〈0.01), and serum IL- 10 contents of the three treatment groups were obviously up-regulated (P〈0.01). The effect of the EA group was apparently superior to that of the medication group in down-regulating lung index (P〈0.05). No significant differences were found among thethree treatment groups in the levels of serum TNF-α and IL-10, and pulmonary TNF-α and IL-10 (P〈0.05). In addition, results of H.E. stain showed that the treatment of medication, moxibustion and EA might reduce H1Nl-induced pulmonary pathological chan- ges. Conclusien EA stimulation of "Feishu" (BL13) can down-regulate lung index and serum TNF-α and up-regulate serum IL- 10 level in viral pneumonia mice, which may contribute to its effect in relieving asthma.
