摘要
以80%乙醇为提取溶剂,以树头菜(Crateva unilocalaris Buch.)黄酮提取率为指标,在单因素试验基础上,用响应面法优化野生树头菜黄酮的提取条件,并测定其体外抗氧化活性。结果表明,最佳提取工艺为提取温度65℃、提取时间1.3 h、液料比28∶1(mL∶g)、提取2次,树头菜黄酮提取率为0.497%。树头菜黄酮具有较强的清除超氧阴离子自由基、羟自由基的能力和还原能力,清除超氧阴离子自由基和羟自由基的IC50分别为0.615、0.415 mg/mL,其抗氧化能力高于抗坏血酸(VC)和叔丁基对苯二酚(TBHQ)。
Using 80% ethanol as extraction solvent,the extraction ratio of flavonoids from Crateva unilocalaris Buch.as index,response surface methodology was used to optimize the extraction of flavonoids from Crateva unilocalaris Buch.with single factor experiment.The antioxidant activity in vitro was evaluated by total reducing power assay.The results showed that the optimal extraction conditions were extraction temperature of 65 ℃,extraction time of 1.3 h,liquid-to-material ratio of 28∶1 (mL∶g) and repeated extraction number of 2.Under these conditions,the extraction ratio of flavonoids was up to 0.497%.Crateva unilocalaris Buch.flavonoids had excellent reducing power and high abilities to scavenge superoxide anion and hydroxyl free radicals with IC50 values of 0.615 mg/mL and 0.415 mg/mL,respectively.The antioxidant effect of flavonoids from Crateva unilocalaris Buch.were higher than that of VC and TBHQ.
