斑马鱼毒性检测方法的优化及应用研究

摘要 目的:通过优化斑马鱼急性毒性检测方法,检测和分析辅酶Q、低聚果糖、双酚A对斑马鱼的毒性作用,探讨其在食品安全性检测与评价中的应用前景。方法优化试验用斑马鱼的基本条件、非水溶性和水中溶解度较低的受试物的处理要求和斑马鱼急性毒性试验条件,观察和记录斑马鱼在不同浓度的辅酶Q、低聚果糖、双酚A的试验溶液中暴露48h的毒性反应和死亡情况,检测和分析不同浓度的辅酶Q、低聚果糖、双酚A对斑马鱼的毒性作用。结果斑马鱼在剂量为100~1000mg/L的辅酶Q试验溶液中饲养48h,未发现斑马鱼有任何毒性反应,辅酶Q对斑马鱼的48hLC50为大于1000mg/L;斑马鱼在剂量为100~1000mg/L的低聚果糖试验溶液中饲养48h,未发现斑马鱼有任何毒性反应,低聚果糖对斑马鱼的48hLC50为大于1000mg/L;斑马鱼在剂量为10mg/L的双酚A试验溶液饲养3h后开始出现毒性反应,在剂量为8mg/L的双酚A试验溶液饲养6h后开始出现毒性反应,在剂量为6mg/L的双酚A试验溶液饲养12h后开始出现毒性反应,双酚A对斑马鱼的48hLC50为7.15mg/L。结论本研究显示优化的斑马鱼急性毒性检测方法能够快速检测食品和食品污染物的急性毒性,其在食品安全性检测与评价中将有较好应用前景。 Objective By optimizing the acute toxicity test method of zebrafish, the toxic effect of coenzyme Q, fructooligosac-charides (FOS) and bisphenol A on zebrafish were detected and analyzed , and to explore the application prospect of the method in food safety test and evaluation. Methods The basic state of zebrafish and the acute toxicity test conditions were optimized. Ze-brafish were exposured to the solution containing different concentrations of coenzyme Q, FOS and bisphenol A for 48h, the ze-brafish toxicity and mortality rate were observed, the toxic effect were detected. Results When zebrafish were fed in the test solu-tion containing 100-1000mg/L coenzyme Q for 48h, no toxicity was found, the LC50 of coenzyme Q in 48h was greater than 1000mg/L. While zebrafish were fed in the test solution containing 100-1000mg/L FOS for 48h, no toxicity was found, the LC50 of FOS in 48h was greater than 1000mg/L. After zebrafish were fed in the test solution containing 10mg/L bisphenol A for 3h, con-taining 8mg/L bisphenol A for 6h,containing 6mg/L bisphenol A for 12h,the toxic reaction could be detected, the LC50 of bisphe-nol A in 48h was 7.15mg/L. Conclusion This study showed that the optimized acute toxicity test method of zebrafish can be used as rapid detection method for the acute toxicity of food and food contaminants, and has a wide range of applications in the safety test and evaluation of food.
