摘要
目的探讨外周血维甲酸相关孤儿受体γt(RORγt)及叉头框P3蛋白(Foxp3)mRNA、血清白细胞介素10(IL-10)、IL-6、IL-17的浓度,在大鼠脊髓缺血再灌注损伤后各时程的演变。方法雄性Wistar大鼠50只,制备大鼠脊髓缺血再灌注损伤模型,据取材时间分为4组(术后1 d、3 d、5 d、7 d与对照组,n=10)。采用实时荧光定量聚合酶链反应(RT-PCR)检测外周血RORγt和Foxp3 mRNA的表达和酶联免疫吸附测定(ELISA)检测血清白细胞介素10(IL-10)、IL-6、IL-17的浓度。结果大鼠脊髓缺血再灌注损伤不同时间组外周血RORγt及Foxp3mRNA表达水平及血清IL-10、IL-6、IL-17与对照组比较,差异具有统计学意义(P<0.05或P<0.01)。结论Thl7/调节性T细胞的失衡与大鼠脊髓缺血再灌注损伤后的发生发展密切相关。
Objective This study aimed to investigate the changes of T-helper 17( Th17) and regulatory T( Treg) cells with rat spinal cord ischemia / reperfusion injury by determining the mRNA expression of retinoid-related orphan receptorγt( RORγt) and forkhead box P3 protein( Foxp3),and to observe the correlation with the status of inflammation. Methods Fifty adult Wistar rat( 380 ~ 420g) were randomly divided into five groups,the control group( n = 10) and four operation groups( n =40). The SCⅡ model was established with a self-made device in the operation group. The segments of ischemia / reperfusion spinal cord were properly taken at interval of 1、3、5 and 7 d after reperfusion. The expression levels of plasma RORγt mRNA and Foxp3 mRNA were detected by real-time quantitative PCR( qRT-PCR). IL-10、IL-6 and IL-17 levels in plasma detected by ELISA.Results Expression of RORγt mRNA and Foxp3 mRNA and level of IL-10、IL-6 and IL-17 were significantly increased in the operation group compared with the control group,with statistical differences( P 0. 05 or P 0. 01). Conclusion The disequilibrium of Th17 and Treg cells has a close relationship with rat spinal cord ischemia / reperfusion injury.
